云南部分地区牛羊体表寄生蜱的形态及分子鉴定

Morphological and molecular identification of parasitic ticks on cattle and sheep in some parts of Yunnan Province

摘要 为了解云南部分地区牛羊体表寄生蜱的种类,对云南6个地区采集的445只蜱的形态进行观察,并采用PCR技术对虫体的细胞色素C氧化酶亚基Ⅰ(coxⅠ)基因序列进行扩增,于NCBI-BLAST在线比对有效序列,利用MEGA 6.0软件构建系统进化树。结果显示:经形态学特征初步鉴定,采集的蜱属为扇头蜱属(Rhipicephalus)。coxⅠ基因序列比对分析显示,研究中所获分离株与微小扇头蜱(Rhipicephalus microplus)同源性达100%,系统进化分析显示分离株与微小扇头蜱处于同一分支。本研究可为云南牛羊体表寄生蜱的种类鉴定和防治提供参考。 In order to explore the species of ticks which parasitized on cattle and sheep in some areas of Yunnan Province,the morphologies of ticks collected in six areas were observed,and the mitochondrial cytochrome coxidase subunitⅠ(coxⅠ)genes of the ticks were amplified by PCR.Then,the effective sequences were subjected to NCBI-BLAST for online alignment,and MEGA 60 was used to construct a phylo-genetic tree.The results showed that,according to the morphological characteristics,the species of the collected ticks was preliminarily iden-tified as Rhipicephalus.The comparative analysis of the coxⅠgene sequence showed that the homology of the gene with Rhipicephalus micro-plus was 100%.The phylogenetic analysis showed that the isolates and Rhipicephalus microplus were in the same branch.The present results provided reference for identification of parasitic tick species on the body surface of cattle and sheep in Yunnan Province and for prevention and control of tick diseases.
