外用制剂“甲节减”对昆明小鼠抗炎镇痛作用的初步探究

Anti-Inflammatory and Analgesic Effects of External Application of 'Jia Jie Jian' on KM Mice

目的:研究中药复方“甲节减”外用制剂的抗炎、镇痛作用并初步探究其抗炎作用机制。方法:将昆明小鼠按体质量随机分为模型组,阳性药组(云南白药酊剂),“甲节减”高、中、低剂量组(20、10、5 mL/kg),每组10只。模型组涂抹空白基质蒸馏水,其余组小鼠外涂相应药物,1次/d,共7 d。采用小鼠急、慢性炎症模型检测药物的抗炎疗效,光电甩尾法检测药物的镇痛作用,ELISA法检测血清肿瘤坏死因子-α(tumor necrosis factor,TNF-α)、白介素-1β(interleukin-1β,IL-1β)含量。结果:小鼠急性耳肿胀实验中,各组小鼠耳廓质量比较,差异无统计学意义(P>0.05);小鼠肉芽肿慢性炎症实验中,与模型组比较,“甲节减”中、高剂量组小鼠肉芽肿质量极显著降低(P<0.01),血清IL-1β含量极显著降低(P<0.01),但各组TNF-α水平差异不显著(P>0.05);小鼠镇痛实验中,与模型组比较,给药后2 h,“甲节减”中、高剂量组小鼠痛阈显著升高(P<0.05,P<0.01)。结论:外用制剂“甲节减”具有抗炎、镇痛作用,其机制可能与降低体内炎症因子含量有关。

Objective:To study the anti-inflammatory and analgesic effects of the external preparation of compound formula‘Jia Jie Jian’and to explore its anti-inflammatory mechanism.Methods:KM mice were randomly divided into the model group,the positive drug group(Yunnan Baiyao Tincture),the Jia Jie Jian groups of high-dose(20 mL/kg),medium-dose(10 mL/kg)and low-dose(5 mL/kg),with 10 mice in each group.The model group was smeared with distilled water,and the other groups was smeared with corresponding medication,once per day for seven days.The anti-inflammatory effect of the drugs was tested with acute and chronic inflammation models in mice,and the analgesic effect of the medications was tested by photoelectric tail flick method,and the serum contents of TNF-αand IL-1βwere detected by ELISA method.Results:In terms of acute ear swelling experiment,there was no statistical difference in the quality of auricle in each group after the treatment(P>0.05).In terms of chronic granuloma inflammation experiment,compared with those in the model group,the dry mass of granuloma were significantly decreased(P<0.01),and the serum content of IL-1βwas significantly reduced in the Jia Jie Jian groups of medium-dose and high-dose(P<0.01);there was no statistical difference in TNF-αamong each group(P>0.05).In terms of analgesic experiment,compared with that in the model group,the pain threshold was significantly increased 2 h after medication in the Jia Jie Jian groups of medium-dose and high-dose(P<0.05,P<0.01).Conclusion:The external preparation of Jia Jia Jian has anti-inflammatory and analgesic effects,which may be related to reducing the content of inflammatory factors in the body.
