桑黄子提取物对荷瘤小鼠免疫功能的影响 被引量:3

摘要 目的 探讨白桦树桑黄子实体提取物(EBPI)对荷瘤小鼠瘤体质量、Caspase-3蛋白表达、免疫功能的影响,并阐明其潜在的作用机制.方法 建立S_(180)荷瘤小鼠模型,即在无菌工作台上将0.2 mL的细胞悬液于30 min内接种每只小鼠右腋窝皮下.将荷瘤小鼠随机分为正常对照组、模型组、环磷酰胺组(CTX)、低0.5 g/(kg·d)、中1 g/(kg·d)和高2 g/(kg·d)剂量EBPI组,每组10只.采用称重法测量计算并观察EBPI对S_(180)荷瘤小鼠免疫器官指数(脾脏指数、胸腺指数、肝脏指数)、各组小鼠瘤体质量的影响;脾脏淋巴细胞计数及Western blotting法观察EBPI对S_(180)荷瘤小鼠脾淋巴细胞及瘤体Caspase-3蛋白表达的影响.结果 低、中和高剂量EBPI组与模型组、CTX组比较脾指数和胸腺指数均提高显著(P<0.01),而肝脏指数无明显升高或降低(P>0.05);与CTX组比较,S_(180)荷瘤小鼠脾淋巴细胞数升高(P<0.05);低、中和高剂量EBPI组瘤体质量及抑瘤率与模型组比较差异具有统计学意义(P<0.01,P<0.05).中、高剂量EBPI组瘤体质量和抑瘤率明显低于CTX组(P<0.05).Western blotting结果显示,低、中和高剂量EBPI组Caspase-3蛋白的表达较对照组明显升高(P<0.05),且Caspase3蛋白的表达量与EBPI浓度呈正相关关系.结论 EBPI可显著提高S_(180)荷瘤小鼠的免疫器官指数,提高脾脏淋巴细胞数,增加Caspase-3蛋白表达,抑制S_(180)荷瘤小鼠瘤体的生长. Objective To investigate the effects of extract from the fruit body of Betula platyphylla and Phellinus igniarius(EBPI) on tumor weight,Caspase-3 protein expression and immune function of tumor bearing mice,and to elucidate its potential mechanism.Method S_(180) tumor bearing mice model was established.Cell suspension(0.2 mL) was inoculated subcutaneously in the right armpit of mice within 30 min on the sterile ultra clean table.The mice were randomly divided into normal control group,model group,cyclophosphamide group(CTX),low 0.5 g/(kg·d),medium 1 g/(kg·d),high 2 g/(kg·d) EBPI groups,with 10 mice in each group.The immune organ index(spleen index,thymus index,liver index) and the number of spleen lymphocytes were measured by weighting method.The tumor weight was measured by weighting method,and the tumor inhibition rate was calculated.Western blotting was used to detect the expression of Caspase-3 in tumor bearing mice.The effects of EBPI on immune function,spleen lymphocyte,tumor and Caspase-3 expression were observed.Results Compared with the model group and CTX group,the spleen index and thymus index of low,medium and high dose EBPI groups were significantly increased(P<0.01),while the liver index was not significantly increased or decreased by EBPI(P<0.05);Compared with CTX group,the number of spleen lymphocytes in S_(180) tumor bearing mice was increased(P<0.05);The tumor weight and tumor inhibition rate of EBPI in three groups were significantly different from those of model group(P<0.01,P<0.05).The tumor weight and tumor inhibition rate in low and high-dose EBPI groups were significantly lower than those in CTX group(P<0.05).Western blotting results showed that the expression of Caspase-3 protein in EBPI group was significantly higher than that in control group(P<0.05),and the expression of Caspase-3 protein was positively correlated with EBPI concentration.Conclusion EBPI can significantly increase the immune organ index,increase the number of spleen lymphocytes,increase the expression of Caspase-3 protein and inhibit the growth of S_(180) tumor bearing mice.
