摘要
目的 通过代谢组学检测技术,探索心火脾虚证型特应性皮炎患者肠道菌群差异共代谢产物,并进行相关分析。方法 研究纳入心火脾虚证型特应性皮炎受试者与健康志愿者各20例。将采集到的粪便标本进行代谢组学检测,得到样本中所有代谢产物及相对含量,探索两组差异代谢产物。利用基因组百科全书(KEGG)数据库进行注释和功能富集分析,再利用人类代谢组数据库(HMDB)进行疾病关联分析。结果 共筛选出心火脾虚证型特应性皮炎患者显著差异代谢产物77个。在KEGG通路富集结果中,胆汁分泌(bile secretion)通路富集最为显著。而与特应性皮炎关联疾病中,关系最为密切的是消化系统疾病,以结直肠癌、溃疡性结肠炎、嗜酸性食管炎、克罗恩病为主。结论 心火脾虚证型特应性皮炎患者肠道菌群共代谢产物呈现出与健康志愿者不一样的代谢轮廓。
Objective To explore the use of metabolomics assay methodologies to identify various co-metabolites of intestinal flora in individuals with evidence of atopic dermatitis in the form of syndrome of heart fire and spleen deficiency.Methods There were 20 healthy volunteers and 20 participants who had syndrome of heart fire and spleen deficiency of atopic dermatitis in this study.To get all metabolites and their relative contents in the samples,as well as to investigate the different metabolites between the two groups,the obtained stool samples were put through metabolomic tests.The kyoto Encyclopedia of Genes and Genomes(KEGG)database was used for annotation and functional enrichment analysis,and the Human Metabolome Database(HMDB) was for disease association analysis.Results Patients with atopic dermatitis in the form of syndrome of heart fire and spleen deficiency were included in the trial,and 77 substantially distinct metabolites were analysed.Bile secretion(choleresis) route was the most significantly enriched KEGG pathway among the results.The digestive system disorders,particularly colorectal cancer,ulcerative colitis,eosinophilic esophagitis,and Crohn′s disease,were the diseases that were most closely related to atopic dermatitis.Conclusion The metabolic profile of intestinal flora co-metabolites in atopic dermatitis in the form of syndrome of heart fire and spleen deficiency is different from that of healthy volunteers.
