蚓激酶对哮喘小鼠气道炎症和TLR2/MyD88/NF-κB信号通路的影响

Effects of lumbrokinase on airway inflammation and TLR2/MyD88/NF-κB signaling pathway in asthmatic mice

摘要 目的研究蚓激酶对过敏性哮喘小鼠TLR2/MyD88/NF-κB信号通路和炎症相关因子IL-5、IL-13、IL-33的影响,探讨蚓激酶对哮喘小鼠气道炎症的作用机制。方法24只SPF级BALB/c雄性小鼠随机分为4组:空白组、模型组、地塞米松组和蚓激酶组,每组6只。HE染色法观察肺组织病理学改变;ELISA法检测肺泡灌洗液(BALF)中IL-5、IL-13、IL-33的表达水平;RT-PCR检测肺组织中TLR2、MyD88、TAK1、IKBα、p65 mRNA水平;Western blot检测肺组织中MyD88、TAK1、IKBα、p65蛋白水平。结果蚓激酶可以改善哮喘小鼠模型炎症细胞浸润,降低BALF中炎症因子IL-5、IL-13、IL-33的表达,以及程度上抑制TLR2/MyD88/NF-κB信号通路相关蛋白和mRNA的激活表达水平。结论蚓激酶对OVA哮喘小鼠气道炎症有明显的改善作用,相关机制可能与TLR2/MyD88/NF-κB信号通路的程度激活抑制有关。 Objective To study the effect of lumbrokinase on TLR2/MyD88/NF-κB signaling pathway and inflammatory related factors IL-5,IL-13 and IL-33 in allergic asthmatic mice,and to explore the mechanism of lumbrokinase on airway inflammation in asthmatic mice.Methods Twenty-four SPF BALB/c male mice were randomly divided into 4 groups:blank group,model group,dexamethasone group and lumbrokinase group,with 6 mice in each group.The pathological changes of lung tissue were observed by HE staining.The expression levels of IL-5,IL-13 and IL-33 in bronchoalveolar lavage fluid(BALF)were detected by ELISA.The mRNA levels of TLR2,MyD88,TAK1,IKBαand p65 in lung tissue were detected by RT-PCR.The protein levels of MyD88,TAK1,IKBαand p65 in lung tissue were detected by Western blot.Results Lumbrokinase could improve the infiltration of inflammatory cells in asthmatic mice,reduce the expression of inflammatory factors IL-5,IL-13 and IL-33 in BALF,and inhibit the activation and expression of TLR2/MyD88/NF-κB signaling pathway related proteins and mRNA to a certain extent.Conclusion Lumbrokinase has a significant improvement effect on airway inflammation in OVA asthmatic mice,and the related mechanism may be related to the degree of activation and inhibition of TLR2/MyD88/NF-κB signaling pathway.
