黄芩炭水溶性碳点细胞毒性及对黄芩苷、黄芩素溶解度的影响研究

摘要 目的研究黄芩炭水溶性碳点的细胞毒性及对黄芩苷、黄芩素溶解度的影响。方法通过炒炭法制备黄芩炭,并提取分离纯化黄芩炭水溶性碳点。采用细胞病变法及CCK-8染色法,测定黄芩炭水溶性碳点的细胞毒性,利用高效液相色谱法研究碳点对黄芩苷、黄芩素在水中溶解度的影响变化。结果黄芩炭水溶性碳点对Hep2、RD细胞毒性低,黄芩炭水溶性碳点浓度31.25μg/mL以上时,能够显著提高黄芩苷在水中的溶解度,黄芩炭水溶性碳点浓度62.5μg/mL以上时,能够显著提高黄芩素在水中的溶解度。结论黄芩炭水溶性碳点具有良好的生物安全性,在一定浓度范围内能显著增加黄芩苷、黄芩素在水中的溶解度。 Objective To study the cytotoxicity of water-soluble carbon dots of charred Scutellariae Radix and its effect on solubility of baicalin and baicalein.Methods Charred Scutellariae Radix was prepared by charcoal frying method,and carbon dots were extracted,separated and purified.The cytotoxicity of water-soluble carbon dots was determined by cytopathic method and CCK-8 staining method.The solubility of baicalin and baicalein in water was determined by HPLC.Results The water-soluble carbon dots had low toxicity to Hep2 and RD cells.The solubility of baicalin in water was significantly increased when the concentration of water-soluble carbon dots was above 31.25μg/mL,and the solubility of baicalein in water was significantly increased when the concentration of water-soluble carbon dots was above 62.5μg/mL.Conclusion The water-soluble carbon dots of charred Scutellariae Radix have good biocompatibility and can significantly increase the solubility of baicalin and baicalein in water within a certain concentration range.
