红甜菜和糖甜菜块根中萜类代谢物的比较分析

摘要 目前对于甜菜生物体内萜类次生代谢产物筛查鉴定的研究偏少,且缺乏系统性的分析。为了探究红甜菜和糖甜菜块根中萜类成分的差异,本研究采用UPLC-MS/MS检测平台和Metware自建数据库对2种甜菜块根中的萜物质进行检测和分析,明确其代谢物质成分及差异,并利用多元统计分析方法从检测到的萜类代谢物数据中筛选差异代谢物。2种甜菜块根共检测出6种类型48种萜类物质,包括13种倍半萜,7种单萜,13种二萜,7种三萜,7种三萜皂苷,以及布卢门醇C;差异代谢物6种,其中地肤子皂苷Ic、齐墩果酸-3-O-木糖基(1→3)葡萄糖醛酸苷、去氢木香内酯和30-去甲常春藤皂苷元在红甜菜中含量较高,女贞苦苷和异鸡蛋花素在糖甜菜中含量较高。根据代谢物Log2FC判定差异倍数较大的成分主要是地肤子皂苷Ic、齐墩果酸-3-O-木糖基(1→3)葡萄糖醛酸苷、去氢木香内酯、30-去甲常春藤皂苷元、异鸡蛋花素。2种甜菜材料中萜类成分存在明显差异,可作为特征物质进一步分析。 At present,there are few studies on the screening and identification of terpenoid secondary metabolites in sugar beet organisms,and there is a lack of systematic analysis.To investigate the differences of terpenoids composition between red beet and sugar beet roots,the UPLC-MS/MS detection platform and metware self-built database were used to detect and analyze terpenes substances in roots of two types of sugar beet,and clarify their metabolite composition and differences,and screen differential metabolites from the detected terpenoid metabolite data using multivariate statistical analysis.A total of 48 types of terpenoids substances were detected in the roots of two types of sugar beet,including 13 sesquiterpenes,7 monoterpenoids,13 diterpenes,7 triterpenes,7 triterpenoid saponins and blue methanol C.Six differential metabolites were identified,among them,Momordin Ic,Oleanolic acid-3-O-xylosyl(1→3)glucuronide,Dehydrocostuslactone and 30-Norhederagenin performed higher content in red beet,and Nuezhengalaside and Isoplumericin performed higher content in sugar beet.According to the Log2FC of metabolites,the main components with larger differential ratios were Momordin Ic,Oleanolic acid-3-O-xylosyl(1→3)glucuronide,30-Norhederagenin and Isoplumericin.There were significant differences in terpenoid composition between sugar beet and red beet,which can be used as characteristic substances for further analysis.
