沉默CpABCG6基因影响苹果蠹蛾对高效氯氰菊酯敏感性

摘要 利用RNAi技术对苹果蠹蛾(Cydia pomonella)CpABCG6基因进行沉默处理,明确了沉默苹果蠹蛾CpABCG6基因可以影响其对高效氯氰菊酯的敏感性。本试验使用饲喂法将苹果蠹蛾的小干扰RNA(siRNA)导入苹果蠹蛾体内,通过qPCR检测CpABCG6基因相对表达量的变化情况。利用浸果法检测沉默CpABCG6基因后苹果蠹蛾对高效氯氰菊酯的敏感性。结果表明,高效氯氰菊酯可显著增加苹果蠹蛾死亡情况,并且CpABCG6基因表达量随着高效氯氰菊酯浓度的增加而呈下降趋势。高效氯氰菊酯亚致死浓度LC_(10)、LC_(50)处理后,CpABCG6基因相对表达量分别为对照组的3.53倍和1.36倍。利用饲喂siRNA干扰效率达到47.96%,之后利用高效氯氰菊酯亚致死浓度LC_(10)、LC_(30)、LC_(50)处理苹果蠹蛾后死亡率增加14.3%、7.2%和7.0%。沉默CpABCG6基因后可增加苹果蠹蛾对高效氯氰菊酯敏感性,进而提升苹果蠹蛾死亡率。 To clarify the silencing of the CpABCG6 gene of codling moth(Cydia pomonella)could affect its sensitivity to high-efficiency cypermethrin by using RNAi technology to silence the CpABCG6 gene of codling moth.In this study,small interfering RNA(siRNA)of silverfish moth was introduced into the silverfish moth by feeding method,and the relative expression of CpABCG6 gene was detected by qPCR.The sensitivity of codling moth to cypermethrin after silencing CpABCG6 gene was detected by fruit dipping.The results showed that cypermethrin could significantly increase the mortality of codling moth,and the expression of CpABCG6 gene decreased with the increase of cypermethrin concentration.The relative expression of CpABCG6 gene after LC_(10)and LC_(50)treatment was 3.53 times and 1.36 times those of the control group,respectively.The interference efficiency of feeding siRNA reached 47.96%.After the treatment of cypermethrin LC_(10),LC_(30)and LC_(50),the mortality rate increased by 14.3%,7.2%and 7.0%.In summa-ry,silencing the CpABCG6 gene increased the sensitivity of silverfish moth to high-efficiency cypermethrin,thereby increasing the mortality rate of silverfish moth.
