摘要
选取多种卤代咔唑,以小鼠肝癌细胞系为测试体系,应用基于瞬时转染的荧光素酶报告基因方法对其AHR激活效力进行比较.发现其在1μM浓度下的激活效力排序为:2367CCZ>136BCZ>27BCZ>1368BCZ>其他,同系物/不同卤素取代之间具有明显差异.因此,基于瞬时转染的报告基因方法可以有效开展类二噁英化合物的毒性评估,且提示可以通过变换转染细胞系开展其他物种特异性的毒性评估.
In this study,a variety of polyhalogenated carbazoles were selected,and their AHR activation potential was evaluated by transient transfection-based luciferase reporter gene assay in mouse hepatoma cells.It was found that the order of activation at 1μM concentration was 2367CCZ>136BCZ>27BCZ>1368BCZ>others,and there were differences among congeners/different halogen substitutions.In summary,the transient transfection-based reporter gene assay can effectively evaluate the potency/toxicity of dioxin-like compounds,and it also suggests that,by using cell lines from other species,performing a species-specific toxicity evaluation could be available.
