hsa_circ_0006404、miR-151-3p在胰腺癌组织中表达及其临床诊断价值

摘要 目的探讨胰腺癌组织中hsa_circ_0006404(circFOXO3)、微小RNA(mir)-151-3p表达水平及其与临床病理特征的关系,以及血清circFOXO3、miR-151-3p诊断胰腺癌的效能。方法以行手术治疗的100例胰腺癌患者为PC组,对照组为90例体检健康者。采用qRT-PCR技术检测癌组织及血清中circFOXO3、miR-151-3p表达变化,并分析与其临床病理特征的关系;比较血清miR-151-3p、circFOXO3诊断胰腺癌的效能。Pearson法分析circFOXO3与miR-151-3p的相关性。结果癌组织中circFOXO3表达水平(0.72±0.14)显著低于癌旁组织(1.01±0.17),miR-151-3p表达水平(1.89±0.36)显著高于癌旁组织(1.03±0.15)(P<0.05);低分化、Ⅲ期、淋巴结转移的胰腺癌癌组织circFOXO3表达水平低于中高分化、Ⅰ+Ⅱ期、无淋巴结转移者,而miR-151-3p表达水平高于中高分化、Ⅰ+Ⅱ期、无淋巴结转移者(P<0.05)。PC组血清circFOXO3表达水平(0.76±0.12)低于对照组(0.98±0.17),miR-151-3p表达水平(1.60±0.34)高于对照组(1.02±0.20)(P<0.05);癌组织和血清中circFOXO3与miR-151-3p表达均呈负相关(γ=-0.619、-0.610,P均<0.001)。circFOXO3、miR-151-3p诊断胰腺癌的曲线下面积分别为0.879、0.937,circFOXO3联合miR-151-3p诊断的曲线下面积为0.968。结论胰腺癌组织及血清中circFOXO3表达水平降低、miR-151-3p表达水平升高,circFOXO3、miR-151-3p表达与部分临床病理特征有关,血清circFOXO3联合miR-151-3p诊断胰腺癌的效能较佳。 Objective To investigate the relationship between the expression levels of hsa_circ_0006404(circFOXO3)and microRNA(mir)-151-3p in pancreatic cancer and clinicopathological characteristics,and the efficacy of serum circFOXO3 and miR-151-3p in the diagnosis of pancreatic cancer.Methods 100 pancreatic cancer patients who underwent surgical treatment were selected as the PC group,and the control group included 90 healthy individuals.The expression changes of circFOXO3 and miR-151-3p in cancer tissues and serum were measured by qRT-PCR,and their relationship with clinicopathological characteristics were analyzed.To compare the diagnostic efficacy of serum miR-151-3p and circFOXO3 in pancreatic cancer.The Pearson method was used to analyze the correlation between circFOXO3 and miR-151-3p.Results The expression level of circFOXO3 in cancer tissue(0.72±0.14)was significantly lower than that in paracancerous tissue(1.01±0.17),and the expression level of miR-151-3p(1.89±0.36)was significantly higher than that in paracancerous tissue(1.03±0.15)(P<0.05).The expression level of circFOXO3 in pancreatic cancer tissues of patients with poorly differentiated,stageⅢ,and lymph node metastasis was lower than that of patients with moderately and well differentiated,stageⅠ+Ⅱ,and no lymph node metastasis,while the expression level of miR-151-3p was higher than that of patients with moderate and high differentiation,stageⅠ+Ⅱ,and no lymph node metastasis(P<0.05).The expression level of serum circFOXO3 in the PC group(0.76±0.12)was lower than that in the control group(0.98±0.17),and the expression level of miR-151-3p in the PC group(1.60±0.34)was higher than that in the control group(1.02±0.20)(P<0.05).There was a negative correlation between circFOXO3 and miR-151-3p in cancer tissue and serum(γ=-0.619,-0.610,all P<0.001).The area under the curve of circFOXO3 and miR-151-3p in the diagnosis of pancreatic cancer was 0.879 and 0.937,and the area under the curve of circFOXO3 combined with miR-151-3p was 0.968.Conclusion The expression of circFOXO3 decreased and miR-151-3p increased in pancreatic cancer tissue and serum.The expressions of circFOXO3 and miR-151-3p are related to some clinicopathological features.Serum circFOXO3 combined with miR-151-3p showed better efficacy in the diagnosis of pancreatic cancer.
