产妇康煎剂对孕期大鼠胎盘的影响

Effect of Chanfukang Decoction on Placenta of Pregnant Rats

目的探讨产妇康煎剂对妊娠大鼠胎盘滋养细胞Bcl-2和Bax表达的影响。方法将36只孕鼠随机分为中药组、生理盐水组和空白对照组。中药组和生理盐水组分别于妊娠第6~19天给予产妇康煎剂和生理盐水灌胃,空白对照组不处理,记录3组大鼠胎盘数量,检测孕鼠外周血人绒毛膜促性腺激素(HCG)水平、谷丙转氨酶(ALT)和谷草转氨酶(AST)水平,胎盘滋养细胞中Bcl-2和Bax mRNA表达及蛋白表达。结果中药组孕鼠胎盘数量和血HCG值低于生理盐水组和空白对照组,差异有统计学意义(P<0.05)。中药组胎盘组织中Bcl-2 mRNA表达量高于生理盐水组和空白对照组,Bax mRNA表达量低于生理盐水组和空白对照组,差异有统计学意义(P<0.05)。中药组胎盘组织中Bcl-2蛋白表达量低于生理盐水组和空白对照组,Bax蛋白表达量高于生理盐水组和空白对照组,差异有统计学意义。结论产妇康煎剂可抑制孕期大鼠胎盘的生长和发育,其机制可能与上调Bax表达和下调Bcl-2表达有关。

Objective To investigate the effect of Chanfukang decoction on the expression of Bcl-2 and Bax in placental trophoblasts of pregnant rats.Methods 36 pregnant rats were randomly divided into Chinese medicine group,normal saline group and blank control group.The Chinese medicine group and the normal saline group were given Chanfukang decoction and normal saline by gavage from the 6th day to the 19th day of pregnancy respectively,while the blank control group was not treated.The number of rat placentas in the three groups was recorded,the levels of HCG,ALT and AST in the blood of pregnant rats were detected,the expression of Bcl-2 and Bax mRNA in placental trophoblasts was detected by PCR,and the expression of Bcl-2 and Bax protein in placental trophoblasts was detected by Western blot.Results The placental number and blood HCG value of pregnant rats in Chinese medicine group were lower than those in normal saline group and blank control group,and the difference was statistically significant(P<0.05).The expression level of Bcl-2 mRNA in placental tissue of Chinese medicine group was higher than that of normal saline group and blank control group,and the expression level of Bax mRNA was lower than that of normal saline group and blank control group,the difference was statistically significant(P<0.05).The expression of Bcl-2 protein in the placenta of the Chinese medicine group was lower than that of the normal saline group and the blank control group,the expression of Bax protein was higher than that of the normal saline group and the blank control group,and the difference was statistically significant.Conclusion Chanfukang decoction can inhibit the growth and development of placenta in pregnant rats,and its mechanism may be related to the up-regulation of Bax expression and downregulation of Bcl-2 expression.
