摘要
采用精密色差仪获取红芪的颜色值数据,应用液相色谱串联质谱(LC-MS/MS)测定红芪中类胡萝卜素类代谢产物的含量,并采用单因素方差分析、PCA分析、聚类分析、相关性分析、OPLS-DA分析等方法,研究红芪颜色与类胡萝卜素类成分的相关性。结果显示,红芪中有17种类胡萝卜素类成分被检出;棕黄色、红棕色、红褐色与黄棕色红芪样品间颜色值具有显著差异性;红芪颜色值与类胡萝卜素类代谢产物含量间存在关联关系,4种颜色样品间17种类胡萝卜素类成分含量存在极大值与极小值的特征,含量分布具有差异性;4种颜色间的差异性代谢产物有2类(胡萝卜素类与叶黄素类)11种,分别为α-胡萝卜素、β-胡萝卜素、叶黄素二月桂酸酯、叶黄素二肉豆蔻酸酯、紫黄质二丁酸酯、紫黄质肉豆蔻酸酯、花药黄质、玉米黄质、紫黄质、叶黄素、β-隐黄质月桂酸,其是红芪产生颜色的呈色物质之一。棕黄色、红棕色、红褐色与黄棕色红芪样品的呈色原因可能与类胡萝卜素类代谢产物的含量及种类有关。
A precision colorimeter was used to obtain color values,and liquid chromatography tandem mass spectrometry(LC-MS/MS)was applied to determine the contents of carotenoid metabolites in Hedysari Radix.One-way ANOVA,PCA analysis,cluster analysis,correlation analysis,OPLS-DA analysis were also used to study the correlation between color and carotenoids in Hedysari Radix.The results showed that 17 carotenoids were detected,and there were significant differences in color values of brown,reddish brown,auburn and yellow-brown samples.It was also found that there was a correlation between color values and contents of carotenoids.Seventeen carotenoids were characterized by maximum and minimal values between the 4 color samples,and the distribution of contents was differential.Eleven differential metabolites were involved in two categories(carotenoids and luteolin),namelyα-carotene,β-carotene,lutein dilaurate,lutein dimyristate,violaxanthin dibutyrate,violaxanthin myristate,antheraxanthin,zeaxanthin,violaxanthin,lutein,andβ-cryptoxanthin laurate,which is one of the color-presenting substances in the production of color in Hedysari Radix.The reasons for the colouration of brown,reddish brown,auburn and yellow-brown Hedysari Radix samples may be related to the content and type of carotenoid metabolites.
