运输应激对后备种公猪生理生化指标及炎症相关基因表达的影响

Effects of transportation stress onphysiological and biochemical indices and expression of inflammation-related genes in boars

摘要 为研究长途运输对后备种公猪生理生化指标及炎症相关基因mRNA表达的影响,本试验选择10头10月龄的长白后备种公猪运输6 h,分别在运输前、运输结束及运输后72 h进行生理指标测定及采集血液进行生化指标的测定,同时对血清中炎症相关基因mRNA的表达量进行分析。结果显示,在运输结束时后备种公猪的直肠温度和呼吸频率极显著高于运输前及运输后72 h(P<0.01),长途运输引起血清中白细胞数量显著升高(P<0.05),其中淋巴细胞和嗜酸性粒细胞数在运输结束时较运输前增加(P<0.05),运输后72 h嗜酸性粒细胞和嗜碱性粒细胞数目及百分比与运输前差异显著(P<0.05),单核细胞数在运输结束时显著低于运输后72 h(P<0.05),运输结束时中性粒细胞百分比升高(P<0.05)。与运输前相比,运输结束时及运输后72 h血清中GSH-Px含量显著升高(P<0.05),SOD、MDA、T-AOC和COR含量运输结束时显著高于运输后72 h(P<0.05);运输对血清中IL-2含量无显著性影响(P>0.05)。血清中IKKα、P50基因表达量在运输结束时和运输后72 h显著降低(P<0.05),TNF-a基因表达量在运输结束时高于运输前(P<0.05),运输对P65、IL-6基因表达量无显著性影响,IL-10基因表达量在运输结束时显著低于运输前和运输后72 h (P<0.05)。结果表明,长距离运输诱发后备种公猪的应激反应,使其生理状态、机体代谢及免疫系统受到一定程度的损伤,导致炎症发生。 To assess the effect of physiological and biochemical parameters and the expression of inflammation related genes in boars under long-distance transportation,10 Landrace boars of ten months had been transported 6 hours,and we measured physiological responses and biochemical indicators of the blood samples just before the transportation,at the end transportation and after transportation 72 h,respectively.mRNA expression of Inflammation related genes in serum was analyzed.The results showed the rectal temperature and respiratory rate at the end of transportation were significantly higher than those before transportation and 72 h after transportation(P<0.01).The number of white blood cells was significantly increased after long-distance transportation(P<0.05),and the number of lymphocytes and eosinophils were increased at the end of transportation compared with before transportation(P<0.05).The number and percentage of eosinophils and basophils after transportation 72 h were significantly different from those of before transportation(P<0.05),the number of monocytes at the end of transportation was significantly lower than that after transportation 72 h(P<0.05),and the percentage of neutrophils was increased at the end of transportation(P<0.05).Compared with before transportation,GSH-Px concentration of serum was significantly increased at the end of transportation and after transportation 72h(P<0.05),SOD,MDA,T-AOC and COR concentration at the end of transportation were significantly higher than those of after transportation 72h(P<0.05),and IL-2 concentration was not affected by transportation(P>0.05).The expression of IKKαand P50 genes in serum were significantly decreased(P<0.05),and mRNA expression of TNF-α at the end of transportation were higher than those of before transportation(P<0.05).Expression of P65 and IL-6 was not affected and expression of IL-10 was significantly reduced at the end of transportation(P<0.05).The results indicated that the long-distance transportation induced stress response of the boar,and the physiological reponses,metabolism and immune system were damaged and leading to inflammation.
