摘要
为优化隔山消多糖提取工艺,该研究以生物反应器培养的隔山消不定根为实验材料进行闪式提取,并以多糖得率为指标,利用正交设计法对提取温度、料液比、提取时间进行了优化。结果表明:提取温度为60℃、料液比为1∶40(g∶mL)、提取时间为70 s时,多糖得率最高,达到7.24%。为了将隔山消不定根应用于产品生产中,开展了抗氧化活性评价研究。结果表明:DPPH和ABTS^(+)清除力和铁离子螯合力随提取物浓度的增加而增加,清除DPPH和ABTS^(+)自由基的半数效应浓度分别为0.39和16 mg/mL。该研究结果表明隔山消不定根在抗氧化产品的开发及生产中具有很好的应用价值。
To optimize extraction process of Cynanchum wilfordii polysaccharides,this study used bioreactor-cultured adventitious roots of C.wilfordii as the experimental material to investigate the optimal extraction conditions(i.e.,extraction temperature,material-liquid ratio,and extraction time)of flash extraction using orthogonal design method with polysaccharide yield as the evaluation index.The results showed that the highest polysaccharide yield(7.24%)was obtained at extraction temperature of 60℃,material-liquid ratio of 1∶40(g∶mL),and extraction time of 70 s.The antioxidant activity of the adventitious roots was then evaluated for future application.The results showed that DPPH and ABTS^(+)scavenging rates and Fe 2+chelating rate increased with increasing extract concentrations,with a median effective concentration of 0.39 mg/mL for DPPH scavenging and 16 mg/mL for ABTS^(+)scavenging.This finding indicated that C.wifordii adventitious roots have an application value in the development and production of antioxidant products.
