摘要
为了探索PLC-γ1基因在哈萨克绵羊不同器官和淋巴组织中的表达差异,试验随机选取6月龄健康的哈萨克绵羊雌雄各5只,进行屠宰,采集样本采用实时荧光定量PCR方法对PLC-γ1基因在哈萨克绵羊不同器官(脾脏、肺脏、胰腺、肾脏、心脏、肝脏、睾丸、卵巢、子宫)及淋巴组织(颈前淋巴、肩前淋巴、网胃淋巴、瓣胃淋巴、瘤胃淋巴、肠系淋巴、十二指肠淋巴、腹股沟淋巴、空肠淋巴和回盲淋巴)的表达规律进行研究。结果表明:哈萨克绵羊胰腺PLC-γ1基因的相对表达量极显著高于其他器官(P<0.01),卵巢PLC-γ1基因的相对表达量显著高于肾脏和子宫(P<0.05),其余器官PLC-γ1基因相对表达量均差异不显著(P>0.05);十二指肠淋巴PLC-γ1基因的相对表达量显著高于瓣胃淋巴(P<0.05),瓣胃淋巴PLC-γ1基因的相对表达量极显著高于其他淋巴组织(P<0.01)。说明PLC-γ1基因在哈萨克绵羊各系统器官和淋巴组织中均有表达,并且在胰腺、卵巢、十二指肠淋巴、瓣胃淋巴中的表达明显,对机体免疫、生殖和消化等方面具有重要作用。
In order to explore the different expressions of the PLC-γ1 gene in different organs and lymphoid tissues of Kazakh ovine,five healthy 6-month-old female and 5 male Kazakh ovine were randomly selected for the test.And the expression law of the PLC-γ1 gene was detected by real-time fluorescent quantitative PCR in different organs(spleen,lung,pancreas,kidney,heart,liver,testis,ovary,uterus)and lymphoid tissues(anterior cervical lymph,anterior shoulder lymph,reticulum lymph,omasum lymph,rumen lymph,intestinal lymph,duodenal lymph,inguinal lymph,jejunal lymph and ileocecal lymph)of Kazakh ovine.The results showed that the relative expression of PLC-γ1 gene in the pancreas of Kazakh ovine was significantly higher than that in other organs(P<0.01).The relative expression of PLC-γ1 gene in the ovary was significantly higher than that in the kidney and uterus(P<0.05),and there was no significant difference in other organs(P>0.05).The relative expression of PLC-γ1 gene in duodenal lymph was significantly higher than that in omal lymph(P<0.05),and the relative expression in it was significantly higher than that in other lymphoid tissues(P<0.01).The results indicated that PLC-γ1 gene was expressed in all organs and lymphoid tissues of Kazakh ovine,and was significantly expressed in pancreas,ovary,duodenal lymph,and omasum lymph.PLC-γ1 gene played an important role in immunity,reproduction and digestion.
