大黄代煎液中5种蒽醌类成分的稳定性研究

摘要 目的建立高效液相色谱-二极管阵列检测法(HPLC-DAD)检测大黄代煎液中5种蒽醌类成分(芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚)的含量,考察不同储存状态对其含量的影响。方法采用HPLC-DAD,流动相为0.1%甲酸水-乙腈体系,线性梯度洗脱,流速为1.0 ml/min,色谱柱温度为25℃,进样体积为5μl,测定不同储存温度和时间下大黄代煎液中5种蒽醌类成分的浓度。结果5种蒽醌类成分在相应浓度范围内线性关系良好,其精密度、重复性、稳定性和加样回收率良好;低温储存28 d内药液5种蒽醌类成分基本稳定,常温和高温储存3 d内基本稳定。结论所建立的HPLC-DAD法稳定性、快速、重复性好,可应用于大黄代煎液中5种蒽醌类成分的含量测定;大黄代煎液在冷藏条件下稳定性较好。 Objective To develop a high-performance liquid chromatography-diode array detection(HPLC-DAD)method for the determination of five anthraquinones(aloeemodin,rhein,emodin,chrysophanol and physcion)in rhubarb decoction.Methods A HPLC method was established to investigate the effects of different storage temperatures and time on the concentration of five anthraquinones in rhubarb decoction.Separation was performed with a gradient mobile phase system of acetonitrile-0.1%formic water solution,the flow rate was 1.0 ml/min,the temperature of the column was 25℃,and the injection volume was 5μl.Results Five anthraquinones were successfully separated using this method,with a good linear relationship in the corresponding concentration range.The precision,repeatability,stability and recovery were good.The five anthraquinones were basically stable within 14 days after low-temperature storage,and basically stable within 3 days after normal temperature and high-temperature storage.Conclusion The HPLC-DAD method is stable,rapid and reproducible.It can be used to determine the contents of five anthraquinones in rhubarb decoction.Rhubarb decoction has good stability under cold storage conditions.
