一测多评法同时测定连翘叶红茶中5种指标成分的含量

摘要 以连翘叶红茶为研究对象,建立一测多评法(QAMS)同时测定连翘叶红茶中芦丁、连翘酯苷A、槲皮素、连翘苷和连翘脂素5种指标成分的含量的方法,并进行方法学考察。采用高效液相色谱(HPLC)法,以连翘苷为内参物,计算其与芦丁、连翘酯苷A、槲皮素和连翘脂素的相对校正因子。比较一测多评法和外标法(ESM)测定5种指标成分检测结果的差异。结果表明,5种指标成分在各自质量浓度范围内线性关系良好(相关系数R均>0.999),其检出限为0.45~0.78μg/m L,定量限为1.49~2.59μg/m L,平均加标回收率为98.25%~99.63%,精密度、加标回收率、稳定性试验结果的相对标准偏差(RSD)均<3.0%,表明QAMS法精密度、准确度、稳定性良好。一测多评法(QAMS)和外标法(ESM)测定10批连翘叶红茶中5种成分含量结果之间无显著差异(P>0.05)。QAMS法适用于连翘叶红茶中5种指标成分的同时测定。 Taking Forsythia suspense leaves black tea as research object,a method for simultaneous determination of rutin,forsythoside A,quercetin,forsythin and forsythia in F.suspense leaves black tea by quantitative analysis of multi-components by single marker(QAMS)was established and the methodology was investigated.Using forsythin as the internal reference,the relative correction factors of forsythin with rutin,forsythoside A,quercetin and forsythia were calculated,and the difference of test results for 5 indicator components were compared between quantitative analysis of multi-components by single marker method and external standard method(ESM).The results showed that the linear relationship of 5 indicator components within their respective mass concentration ranges was good(correlation coefficients R>0.999),the detection limit was 0.45-0.78μg/ml,the quantification limit was 1.49-2.59μg/ml,the average standard recovery rates were 98.25%-99.63%,and the relative standard deviation(RSD)of precision,standard recovery rate,and stability test results were all less than 3.0%,indicating that the QAMS method had good precision,accuracy,and stability.There was no significant difference in the determination of 5 components contents in 10 batches of F.suspense leaves black tea by QAMS method and the ESM method(P>0.05).The QAMS method was suitable for the simultaneous determination of 5 indicator components in F.suspense leaves black tea.
