狼疮性肾炎患者外周血单个核细胞自噬相关蛋白表达上调且与肾损害相关

The expression of autophagy-related proteins in peripheral blood mononuclear cells of patients with lupus nephritis is upregulated and related to kidney damage

目的 研究活动性系统性红斑狼疮(SLE)患者狼疮肾炎活动性与自噬及炎症的关系。方法 采用Western blot法检测狼疮肾炎组与非狼疮肾炎组SLE患者外周血单个核细胞(PBMC)自噬相关蛋白微管相关蛋白1轻链3(LC3)和P62的表达,ELISA检测SLE患者血清肿瘤坏死因子α(TNF-α)和γ干扰素(IFN-γ)的水平。采用Pearson法分析LC3Ⅱ/LC3Ⅰ比值与SLE疾病活动度评分(SLEDAI)、尿蛋白、 TNF-α及IFN-γ水平的相关性。结果 SLE患者PBMC的LC3表达增加、 P62表达减少。与不伴肾脏损害的SLE患者相比,伴肾脏损害的SLE患者组自噬水平增加更明显。SLE患者血清TNF-α和IFN-γ水平增加。LC3Ⅱ/LC3Ⅰ比值与SLEDAI(r=0.4560)、 24 h尿蛋白(r=0.3753)、 IFN-γ(r=0.5685)水平正相关,与TNF-α水平(r=0.04 683)无相关性。结论 SLE患者PBMC存在自噬,自噬水平与狼疮肾炎患者肾损害及炎症有一定相关性。

Objective To identify the relationship between nephritis activity,autophagy and inflammation in patients with SLE.Methods Western blot analysis was used to detect the expression of microtubule-associated protein 1 light chain 3(LC3)and P62 in peripheral blood mononuclear cells(PBMCs)of SLE patients with lupus nephritis and non-lupus nephritis patients.Tumor necrosis factorα(TNF-α)and interferon(IFN-)in the serum of SLE patients were determined by ELISA.The correlation between LC3 II/LC3 I ratio and SLE disease activity score(SLEDAI),urinary protein,TNF-αand IFNlevels was analyzed by Pearson method.Results The expression of LC3 was increased and P62 was decreased in SLE patients.TNF-αand IFN-were increased in the serum of SLE patients.LC3 II/LC3 I ratio was positively correlated with SLEDAI(r=0.4560),24 hour urine protein(r=0.3753),IFN-(r=0.5685),but had no correlation with TNF-α(r=0.04683).Conclusion Autophagy is found in PBMCs of SLE,and the autophagy is correlated with renal damage and inflammation in patients with lupus nephritis.
