乌鸡白凤片对大鼠胚胎-胎仔发育的毒性研究

Toxicity of Wuji Baifeng Tablets on Embryo-fetal Development of Rats

目的研究乌鸡白凤片对大鼠胚胎-胎仔发育的毒性,为临床用药提供参考。方法86只孕鼠随机分为乌鸡白凤片2.5,5,10 g/kg剂量组和溶媒对照组,于妊娠第6天至第17天连续重复灌胃给药,定期进行临床观察、体重和摄食量测定;妊娠第20天解剖,解剖后观察带胎子宫、卵巢以及着床、吸收胎、死胎、活胎、黄体等相关指标,并检查活胎外观、内脏和骨骼。结果乌鸡白凤片各剂量组孕鼠的体重、摄食量、生殖器官、生殖功能指标、外观形态、内脏和骨骼发育等相关指标均未见明显异常改变。结论妊娠SD大鼠母体无不良反应剂量及胎仔无不良反应剂量均为10 g/(kg·d)。

Objective To explore the toxicity of Wuji Baifeng Tablets on embryo-fetal development of rats and provide reference for the further clinical usage.Methods Total 86 fertilized rats were randomly divided into solvent control group and Wuji Baifeng Tablets 2.5g/kg,5g/kg,10g/kg groups.The rats were intragastrically administered with Wuji Baifeng Tablets from day 6 of pregnancy(GD 6)once daily until GD 17.The clinical manifestation of animals was observed every day during test period,feed intake and bodyweight were detected regularly.Anatomic examination was performed on GD20.The relative indexes of main organs including fetal uterus,ovary,implantation gland,absorption fetus,stillbirth,live fetus and corpus luteum were observed.The lengths of fetus crown-rump and tail were measured and the appearance,skeleton,and viscera of fetus were detected.Results Compared with solvent control group,the body weight,food intake,reproductive organs,reproductive function,appearance,visceral and skeletal development indexes in Wuji Baifeng Tablets groups had no significant abnormal changes.Conclusion The maternal and fetal doses of pregnant SD rats with no adverse reactions both can be 10 g/(kg·d)
