决明子水提物对急性酒精性肝损伤小鼠的保护作用研究

Protective Effect of Water Extract of Cassiae Semen on Acute Alcoholic Liver Injury in Mice

目的探究决明子水提物对急性酒精性肝损伤小鼠的保护作用。方法将60只ICR小鼠随机分为空白组、模型组、阳性药水飞蓟素组(0.2 g/kg)、决明子水提物低、中、高剂量(2.25,4.5,9 g/kg)组。空白组和模型组灌胃生理盐水,给药组分别灌胃相应剂量的决明子水提物及水飞蓟素溶液,连续给药7 d,除空白组外其余各组末次给药后9 h,灌胃大剂量42°白酒。观察与分析各组小鼠体质量、肝脏指数、油红O染色;检测小鼠血清和肝脏生化指标。结果与模型组相比,决明子水提物中、高剂量组体质量显著增长,肝脏指数显著降低;决明子水提物高剂量组总胆固醇(TC)、甘油三酯(TG)、丙二醛(MDA)、丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、肿瘤坏死因子(TNF-α),白介素1β(IL-1β)和白介素6(IL-6)含量显著降低,谷胱甘肽(GSH)含量升高(P<0.01,P<0.05);病理切片观察结果表明,各给药组小鼠肝脏病理变化中脂肪滴变性明显降低。结论决明子水提物能显著减轻酒精引起的肝损伤,具有保肝的作用。

Objective To explore the protective effect of water extract of Cassiae Semen on acute alcoholic liver injury in mice.Methods Sixty ICR mice were randomly divided into blank group,model group,positive(silymarin)group(0.2 g/kg),and Cassiae Semen water extract low-dose,medium-dose and high-dose(2.25,4.5,9 g/kg)groups.The rats in blank group and model group were gavaged with physiological saline,while the rats in the experimental groups were given corresponding doses of water extract of Cassiae Semen and silymarin solution by gavage,respectively,for 7 days.Except for the blank group,the rats in the other groups were given a large dose of 42-degree Erguotou spirit by gavage at the 9th hour after the last administration.The body weight,liver index and oil Red O staining of each group were observed and analyzed.Serum and liver biochemical indexes of mice were detected.Results Compared with the model group,the mice body weight of Cassiae Semen water extract medium and high dose groups increased significantly,and the liver index decreased significantly.The levels of total cholesterol(TC),triglyceride(TG),malondialdehyde(MDA),alanine aminotransferase(ALT),aspartate aminotransferase(AST),tumor necrosis factor-α(TNF-α),interleukin-1β(interleukin-1β)and interleukin-6(IL-6)in the mice of Cassiae Semen water extract high dose group significantly decreased(P<0.01),while the content of glutathione(GSH)increased(P<0.05).The results of pathological section observation showed that among the liver pathological changes of mice,the fat droplet degeneration in each administration group was significantly reduced.Conclusion The water extract of Cassiae Semen can significantly reduce the liver injury caused by alcohol,and has the function of protecting liver.
