BMP7-Ids信号通路对子宫内膜癌细胞增殖、侵袭的作用

摘要 目的探讨骨成型蛋白7(BMP7)-Ids信号通路对子宫内膜癌细胞增殖、侵袭的作用。方法以人子宫内膜癌细胞Ishikawa作为研究对象,将细胞分为四组,即空白对照组(不加BMP7)、A组(2 ng/ml BMP7)、B组(5 ng/ml BMP7)、C组(10ng/mlBMP7)。采用qRT-PCR法检测分化抑制因子(Id)1、Id3的mRNA;Western blot法检测Id1、Id3蛋白表达情况;划痕实验检测细胞迁移能力;采用MTT法检测细胞增殖;采用Transwell实验观察细胞的侵袭情况。每组试验均重复4次,取平均值。结果A组、B组和C组Id1、Id3的mRNA和蛋白表达水平均高于空白对照组,其中C组指标高于A组、B组,B组指标高于A组,差异均有统计学意义(P<0.05);24、36和48h,A、B和C组细胞增殖率高于空白对照组,C组细胞增殖率高于A组和B组,且B组细胞增殖率高于A组,差异有统计学意义(P<0.05)。A、B和C组细胞侵袭细胞数和划痕相对愈合面积高于空白对照组,差异有统计学意义(P<0.05);C组侵袭细胞数和划痕相对愈合面积高于A组和B组,且B组侵袭细胞数和划痕相对愈合面积高于A组,差异有统计学意义(P<0.05)。结论子宫内膜癌细胞中BMP7-Ids信号通路的表达上调可激活细胞的增殖和侵袭作用,从而促进子宫内膜癌病情的发展。 Objective To investigate the effect of bone morphogenetic protein 7(BMP7)-Ids signaling pathway on the pro-liferation and invasion of endometrial cancer cells.Methods Human endometrial cancer cells Ishikawa were divided into four groups:blank control group(without BMP7),group A(2 ng/ml BMP7),group B(5 ng/ml BMP7)and group C(10 ng/ml BMP7).The mRNA of inhibitor of differentiation(Id)1 and Id3 were detected by qRT-PCR.The expression of Id1 and Id3 protein were detected by Western blot.Cell migration ability was detected by scratch test.Cell proliferation was detected by MTT assay.The cell invasion was observed by Transwell assay.The experiment was repeated 4 times in each group,and the average value was taken.Results The mRNA and protein expression levels of Id1 and Id3 in group A,B and C were higher than those in blank control group,and the indexes of group C were higher than those of group A and B,and the indexes of group B were higher than those of group A,with statistical significances(P<0.05).At 24,36 and 48 h,the cell proliferation rates of group A,B and C were higher than that of blank control group,the cell proliferation rate of group C was higher than those of group A and B,and the cell proliferation rate of group B was higher than that of group A,the differences were statis-tically significant(P<0.05).The number of invasive cells and the relative healing area of scratches in group A,B and C were higher than those in blank control group,the differences were statistically significant(P<0.05).The number of invasive cells and the relative healing area of scratches in group C were higher than those in group A and B,and the number of inva-sive cells and the relative healing area of scratches in group B were higher than those in group A,the differences were statis-tically significant(P<0.05).Conclusion The up-regulated expression of BMP7-Ids signaling pathway in endometrial can-cer cells can activate cell proliferation and invasion,thus promoting the development of endometrial cancer.
