miR-155通过调节ETS1促进鼻咽癌细胞侵袭转移能力的研究 被引量:1

摘要 目的 观察微小RNA(micro RNA)miR-155在鼻咽癌细胞侵袭转移中的作用。方法 以鼻咽癌CEN1和5-8F细胞作为研究对象,分别转染miR-155 NC、miR-155 inhibitor、miR-155 mimics和ETS1 siRNA,运用Transwell实验检测miR-155和ETS1对鼻咽癌细胞迁移和侵袭能力的作用,RT-qPCR检测细胞miR-155表达;蛋白印记实验检测ETS1、MMP2和MMP9蛋白表达。结果 与miR-155 NC组相比,miR-155 inhibitor组细胞侵袭[(237.33±15.63)个vs.(117.00±5.72)个]和迁移[(331.00±9.42)个vs.(262.00±11.05)个]能力均降低(P<0.05),ETS1[1.00 vs.(0.77±0.04)]、MMP2[1.00 vs.(0.58±0.05)]和MMP9[1.00 vs.(0.38±0.06)]蛋白表达均下调(P<0.05);miR-155 mimics组细胞侵袭[(134.00±8.83)个vs.(326.00±12.68)个]和迁移[(236.67±12.39)个vs.(458.00±18.38)个]能力均增强(P<0.05),ETS1[1.00 vs.(1.61±0.06)]、MMP2[1.00 vs.(2.63±0.06)]和MMP9[1.00 vs.(3.38±0.06)]蛋白表达均升高(P<0.05)。与miR-155 mimics组相比,miR-155 mimics+ETS1 siRNA组的细胞迁移[(451.67±22.10)个vs.(229.00±14.45)个]和侵袭[(402.33±8.22)个vs.(229.00±24.91)个]能力均减弱(P<0.05),ETS1[(1.63±0.28)vs.(0.51±0.02)]、MMP2[(2.41±0.02)vs.(1.00±0.07)]、MMP9[(2.37±0.03)vs.(1.02±0.11)]蛋白的表达均下降(P<0.05)。结论 miR-155可能通过调节转录因子ETS1促进细胞外基质降解过程,从而发挥促进鼻咽癌细胞的迁移与侵袭作用。 Objective To observe the role of miR-155 in the invasion and migration of nasopharyngeal carcinoma cells.Methods Nasopharyngeal carcinoma CEN1 and 5-8F cells were transfected with miR-155 NC,miR-155 inhibitor,miR-155 mimics and ETS1 siRNA,respectively.The effects of miR-155 and ETS1 on the migration and invasion of nasopharyngeal carcinoma cells were detected by Transwell assay.RT-qPCR was used to detect the expression of miR-155,and the expression of ETS1,MMP2 and MMP9 proteins were detected by Western blot.Results Compared with miR-155 NC group,the ability of invasion[(237.33±15.63)vs.(117.00±5.72)]and migration[(331.00±9.42)vs.(262.00±11.05)]of cells in miR-155 inhibitor group was decreased(P<0.05),the expression of ETS1[1.00 vs.(0.77±0.04)],MMP2[1.00 vs.(0.58±0.05)]and MMP9[1.00 vs.(0.38±0.06)]protein was decreased(P<0.05);the ability of invasion[(134.00±8.83)vs.(326.00±12.68)]and migration[(236.67±12.39)vs.(458.00±18.38)]of cells in miR-155 mimics group was enhanced(P<0.05),and the expression of ETS1[1.00 vs.(1.61±0.06)],MMP2[1.00 vs.(2.63±0.06)]and MMP9[1.00 vs.(3.38±0.06)]protein was increased(P<0.05).Compared with miR-155 mimics group,the ability of cell migration[(451.67±22.10)vs.(229.00±14.45)]and invasion[(402.33±8.22)vs.(229.00±24.91)]in miR-155 mimics+ETS1 siRNA group was decreased(P<0.05),and the expression of ETS1[(1.63±0.28)vs.(0.51±0.02)],MMP2[(2.41±0.02)vs.(1.00±0.07)],MMP9[(2.37±0.03)vs.(1.02±0.11)]protein was decreased(P<0.05).Conclusion miR-155 may promote the migration and invasion of nasopharyngeal carcinoma cells by regulating ETS1 and promoting the degradation of extracellular matrix.
