当归芍药散对血管性痴呆大鼠炎症因子及神经细胞凋亡的影响

摘要 目的:探讨当归芍药散对血管性痴呆模型大鼠炎症因子、神经细胞凋亡的影响。方法:采用双侧颈动脉永久结扎法制备血管性痴呆模型大鼠,随机分为假手术组、模型组、当归芍药散低剂量组、当归芍药散高剂量组、尼莫地平组。当归芍药散低剂量组给予当归芍药汤1.8 g/kg;当归芍药散高剂量组给予当归芍药汤7.2 g/kg;尼莫地平组给予尼莫地平片20 mg/kg;假手术组、模型组均给予等体积蒸馏水。各组大鼠给药频率为1次/d,持续灌胃4周。采用Morris水迷宫实验计算大鼠逃避潜伏期、穿越平台次数,测定大鼠认知功能;HE染色观察大鼠海马神经元病理变化及神经细胞凋亡情况;Western blotting检测大鼠Bcl-2、Bax蛋白表达;免疫组化染色检测大鼠Bcl-2、Bax平均吸光度值;ELISA测定血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)含量。结果:与假手术组相比,模型组病理学结构疏散,排列紊乱,逃避潜伏期时间延长,穿越平台次数减少。模型组大鼠炎症因子、Bax蛋白表达均高于假手术组,Bcl-2蛋白则较低,差异有统计学意义(均P<0.01)。与模型组相比,当归芍药散各剂量组病理学形态明显改善,逃避潜伏期时间缩短,穿越平台次数增加。当归芍药散各剂量组炎症因子、Bax蛋白均低于模型组,而Bcl-2蛋白则较高,差异有统计学意义(均P<0.01)。结论:当归芍药散对血管性痴呆大鼠具有积极治疗效果,其机制与降低炎症因子、抑制神经细胞凋亡有关。 Objective:To investigate effect of Danggui Shaoyao powder on inflammatory factors and nerve cell apoptosis in rats with vascular dementia.Methods:The rat model of vascular dementia was established by permanent ligation of bilateral carotid arteries.The rats were randomly divided into sham-operated group,model group,low-dose Danggui Shaoyao group,high-dose Danggui Shaoyao group and nimodipine group.The low-dose Danggui Shaoyao powder group were treated with Danggui Shaoyao decoction 1.8 g/kg.The high-dose Danggui Shaoyao powder group were given Danggui Shaoyao decoction 7.2 g/kg.The nimodipine group were given nimodipine tablets 20 mg/kg.The sham-operation group and the model group were given the same volume of distilled water.The administration frequency of each group was once a day for 4 weeks by gavage.Morris water maze test was used to calculate escape latency and number of platform crossings.The cognitive function of rats was measured.The pathological changes of hippocampal neurons and neuronal apoptosis were observed by HE staining.The expression of Bcl-2 and Bax protein were detected by Western blotting.The average Bcl-2 and Bax absorbance were detected by immunohistochemical staining.Serum tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were measured by ELISA.Results:Compared with the sham operation group,the pathological structure of the model group was evacuated and disordered,the escape latency time prolonged,the number of crossing platform reduced.The expressions of inflammatory factors and Bax protein in the model group were higher than those in the sham operation group,while the Bcl-2 protein lower,difference statistically significant(all P<0.05).Compared with the model group,the pathological morphology of each dose group of Danggui Shaoyao powder were significantly improved,the escape latency time shortened,and the number of crossing platform increased.The inflammatory factors and Bax protein in each dose group of Danggui Shaoyao powder were lower than those in the model group,while the Bcl-2 protein higher,difference statistically significant(all P<0.05).Conclusion:Danggui Shaoyao powder has certain therapeutic effect on vascular dementia rats,and its mechanism may be achieved by reducing the expression of inflammatory factors and inhibiting nerve cell apoptosis.
