摘要
目的:检测肺金生方对肺癌干细胞自我更新的抑制作用,并探究其可能的分子机制。方法:采用无血清培养法从人源A549肺癌细胞中诱导肺癌干细胞,建立肺癌干细胞体外培养体系;通过球落形成实验检测肺金生方对肺癌干细胞自我更新的作用;同时,检测肺金生方对肺癌干细胞相关分子造血干细胞抗原CD133、转录因子SOX2、八聚体结合转录因子4(OCT4)及Notch信号通路关键分子Notch1、Jagged1表达的影响。结果:无血清培养法培养的肺癌干细胞成球落状生长,肺癌干细胞表面标志物CD133及干细胞核转录因子SOX2、OCT4的表达增加,表明本研究成功建立了肺癌干细胞体外培养体系。肺金生方可显著减少肺癌干细胞的球落形成数量及体积。与对照组相比,肺金生方可显著抑制肺癌干细胞相关分子CD133、SOX2、OCT4及Notch信号通路关键分子Notch1、Jagged1的表达。结论:肺金生方可抑制肺癌干细胞自我更新,其作用机制可能与抑制Notch1信号通路相关分子有关。
Objective:To detect the effect of Feijinsheng Recipe on the self-renewal of lung cancer stem cells,and to explore its possible molecular mechanism.Methods:Lung cancer stem cells were induced from human A549 lung cancer cells by serum-free culture method,and an in vitro culture system of lung cancer stem cells was established.Expressions of lung cancer stem cell markers CD133,SOX2,OCT4 and Notch signaling pathway key molecules Notch1 and Jagged1were determined.Results:The lung cancer stem cells cultured in se-rum-free culture method grew into spheroids,and the expressions of lung cancer stem cell surface marker CD133 and stem cell nuclear transcription factors SOX2 and OCT4 increased,indicating that the in vitro culture system of lung cancer stem cells was successfully established in this study.After using Feijinsheng Recipe to act on lung cancer stem cells,the spheroid formation experiment showed that Feijinsheng Recipe could significantly inhibit the number and volume of spheroid formation of lung cancer stem cells.Compared with the control group,Feijinsheng Recipe could significantly inhibit the expressions of lung cancer stem cell markers CD133,SOX2,OCT4 and Notch1,Jagged1,key molecules of Notch signaling pathway.Conclusion:Fejinsheng Recipe can inhibit the self-renewal of lung cancer stem cells,and its mechanism may be related to the inhibition of Notch1 signaling pathway-related molecules.
