皮肤鳞状细胞癌组织中Beclin 1、LC3表达与其临床病理特征的关系

Relationship Between Beclin 1 and LC3 in Cutaneous Squamous Cell Carcinoma Tissues and Clinicopathological Characteristics

目的 探究皮肤鳞状细胞癌(CSCC)组织中苄氯素1(Beclin 1)、微管相关蛋白l轻链3(LC3)表达与其临床病理特征的关系。方法 收集CSCC患者的病变组织60份和非癌症患者的正常皮肤组织30份,采用实时荧光定量聚合酶链反应(qRT-PCR)检测组织中Beclin 1、LC3在mRNA水平的表达;Western Blot检测组织中Beclin 1、LC3在蛋白水平的表达,免疫组化法检测组织中Beclin 1、LC3表达。结果 CSCC组的组织Beclin 1、LC3 mRNA表达量均明显低于正常组(P<0.05)。Beclin 1和LC3在正常组织中呈高表达,正常组的蛋白表达量均显著高于CSCC组(P<0.05)。Beclin 1和LC3在CSCC组织中部分阳性表达,随着分化程度越低、病理分级越高、淋巴结发生转移,Beclin 1和LC3的阳性表达率均明显降低(P<0.05)。结论 Beclin 1和LC3表达与CSCC发生发展有关,其表达率降低意味着自噬活性降低,CSCC恶性程度高。

Objective To explore the relationship between beclin 1(Beclin 1)and microtubule-associated protein 1 light chain 3(LC3)in cutaneous squamous cell carcinoma(CSCC)tissues and clinicopathological characteristics.Methods A total of 60 lesion tissue specimens from patients with CSCC and 30 normal skin tissue specimens from non-cancer patients were collected.The mRNA levels of Beclin 1 and LC3 were detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).The expressions of Beclin 1 and LC3 proteins were detected by Western Blot.The expressions of Beclin 1 and LC3 were detected by immunohistochemistry.Results The expression levels of Beclin 1 and LC3 mRNA in CSCC group were significantly lower than those in normal group(P<0.05).The expressions of Beclin 1 and LC3 were up-regulated in normal tissues.The levels of Beclin 1 and LC3 proteins in normal group were significantly higher than those in CSCC group(P<0.05).The expressions of Beclin 1 and LC3 were partially positive in CSCC tissues.The positive expression rates of Beclin 1 and LC3 were significantly decreased in patients with low differentiation,high pathological grading and lymph node metastasis(P<0.05).Conclusion The expressions of Beclin 1 and LC3 are related to the occurrence and development of CSCC.The decrease of their expression rates means the decline of autophagy activity and high malignancy of CSCC.
