摘要
目的探讨piRNA-001184在多发性骨髓瘤(multiple myeloma,MM)中的表达及其与CTGF、CDKN2A基因甲基化的相关性。方法选取2020年8月至2022年12月于内蒙古科技大学包头医学院第一附属医院就诊的69例MM患者纳入实验组,收集同期18例缺铁性贫血患者纳入对照组。应用荧光定量聚合酶链反应检测患者的piRNA-001184表达水平,采用Pearson线性相关分析piRNA-001184与CTGF、CDKN2A表达水平的相关性。结果实验组患者的piRNA-001184表达水平显著高于对照组[(21.56±6.19)vs.(10.60±2.45),t=7.388,P<0.001]。不同病程患者的piRNA-001184表达水平比较差异有统计学意义(F=110.977,P<0.001),表达水平依次为初发组>复发组>缓解组。不同国际分期系统(international staging system,ISS)患者的piRNA-001184表达水平比较差异有统计学意义(P<0.05),表达水平依次为Ⅰ期<Ⅱ期<Ⅲ期。piRNA-001184表达水平与CTGF、CDKN2A基因表达水平均呈正相关(P<0.05)。结论MM患者piRNA-001184表达明显上调。监测piRNA-001184可能对MM的诊断、分期及预后判断有参考价值。
Objective To investigate the expression of piRNA-001184 in multiple myeloma(MM)and its correlation with CTGF,CDKN2A gene methylation.Methods Sixty-nine MM patients treated in the First Affiliated Hospital of Baotou Medical College of Inner Mongolia University of Science and Technology from August 2020 to December 2022 were included in experimental group,and 18 patients with iron deficiency anemia were included in control group during the same period.The expression level of piRNA-001184 was detected by fluorescence quantitative polymerase chain reaction,and the correlation between piRNA-001184 and the expression level of CTGF and CDKN2A was analyzed by Pearson linear correlation analysis.Results The expression level of piRNA-001184 in experimental group was significantly higher than that in control group[(21.56±6.19)vs.(10.60±2.45),t=7.388,P<0.001].There was statistical significance in expression level of piRNA-001184 in patients with different disease course(F=110.977,P<0.001),and the expression level was in initial onset group>relapse group>remission group.The expression level of piRNA-001184 in patients with different international staging system(ISS)was significantly different(P<0.05),and the expression level was in stageⅠ<stageⅡ<stageⅢ.The expression level of piRNA-001184 was positively correlated with that of CTGF and CDKN2A genes(P<0.05).Conclusion The expression of piRNA-001184 was significantly up-regulated in MM patients.Monitoring piRNA-001184 may have reference value for the diagnosis,staging and prognosis of MM.
