摘要
目的通过对鲜地黄、生地黄及熟地黄的抗氧化活性及褐变酶活性进行检测及相关性分析,揭示地黄不同炮制品之间抗氧化活性的差异并为相关机制研究奠定一定的理论基础。方法用1,1-二苯基-2-三硝基苯肼(2,2-diphenyl-1-picrylhydrazyl,DPPH)、2,2’-联氮-双-(3-乙基苯并噻唑啉-6-磺酸)(2,2’-azobis-3-ethylbenzoinesulfonic acid,ABTS)和铁离子还原/抗氧化能力法(ferric reducing antioxidant potential,FRAP)3种体外抗氧化模型对鲜地黄、生地黄及熟地黄的醇提取物和水提取物的抗氧化活性进行比较。同时通过检测多酚氧化酶(polyphenol oxidase,PPO)和过氧化物酶(peroxidase,POD)2种褐变酶的活性来评价鲜地黄、生地黄及熟地黄的酶促褐变反应的活性。结果地黄的不同炮制品均有一定的抗氧化活性,且3种体外抗氧化模型的实验结果基本一致;在水提取物实验中,地黄不同炮制品的抗氧化活性依次为鲜地黄>熟地黄>生地黄;在醇提取物实验中,地黄不同炮制品的抗氧化活性依次为鲜地黄>生地黄>熟地黄。本实验通过对PPO和POD 2种褐变酶的活性进行检测和分析发现,地黄不同炮制品的褐变酶活性差异较大,且PPO和POD 2种褐变酶的活性实验结果基本一致;地黄不同炮制品的PPO和POD 2种褐变酶的活性从高到低依次为生地黄>鲜地黄>熟地黄。结论本文通过对地黄不同炮制品的抗氧化活性与褐变酶活性的综合比较和分析可知,地黄不同炮制品的抗氧化活性与酶促褐变反应有一定的相关性。
Objective Through the detection and correlation analysis of antioxidant activity and browning enzyme activity of fresh,raw and prepared of Rehmannia glutinosa Libosch.radix,this paper aims to reveal the differences of antioxidant activity and related mechanisms between different processed products of Rehmannia glutinosa Libosch.radix.Methods The antioxidant activity of the alcoholic extracts and water extracts of fresh,raw and prepared of Rehmannia glutinosa Libosch.radix were evaluated by 2,2-diphenyl-1-picrylhydrazyl(DPPH),2,2'-azobis-3-ethylbenzoinesulfonic acid(ABTS)and ferric reducing antioxidant potential(FRAP)assay.At the same time,the activity of enzymatic browning reaction of fresh,raw and prepared of Rehmannia glutinosa Libosch.radix was evaluated by detecting the activity of polyphenol oxidase(PPO)and peroxidase(POD).Results Different processed products of Rehmannia glutinosa Libosch.radix showed certain antioxidant activity,and the experimental results of the 3 in vitro antioxidant models were basically same;In the experiment of water extract,the antioxidant activity of different processed products of Rehmannia glutinosa Libosch.radix was as follows:fresh Rehmannia glutinosa Libosch.radix prepared of Rehmannia glutinosa Libosch.radix raw Rehmannia glutinosa Libosch.radix;In the experiment of alcohol extract,the antioxidant activity of different processed products of Rehmannia glutinosa Libosch.radix was as follows:fresh Rehmannia glutinosa Libosch.radix raw Rehmannia glutinosa Libosch.radix prepared of Rehmannia glutinosa Libosch.radix;Through the detection and analysis of the activities of PPO and POD,it was found that the activities of browning enzymes in different processed products of Rehmannia glutinosa Libosch.radix were quite different,and the experimental results of the activities of PPO and POD were basically same;The activities of PPO and POD in different processed products of Rehmannia glutinosa Libosch.radix from high to low was as follows:raw Rehmannia glutinosa Libosch.radix frexh Rehmannia glutinosa Libosch.radix prepared of Rehmannia glutinosa Libosch.radix.Conclusion Through the comprehensive comparative analysis of antioxidant activity and browning enzyme activity of different processed products of Rehmannia glutinosa Libosch.radix,it can be seen that the antioxidant activity of different processed products of Rehmannia glutinosa Libosch.radix has a certain correlation with enzymatic browning reaction.
