足月小样儿与正常足月儿母亲胎盘菌群差异研究

Differences in maternal placental microbiota between full-term small-for-gestational-age infants and normal full-term infants

摘要 目的探讨足月小样儿与正常足月儿母亲胎盘菌群差异,为今后开展深入研究提供数据参考。方法采集2018年1月至9月在昆明医科大学第一附属医院产科规律产检并住院分娩的足月儿母亲的胎盘组织,共9例,按出生体重分为足月小样儿组和正常足月儿组。应用组织DNA提取试剂盒提取胎盘组织菌群DNA,采用16S rDNA高通量测序技术检测样本,分析测序结果,比较两组胎盘菌群差异。结果两组胎盘菌群在门水平上无差异(P>0.05),均以厚壁菌门、变形菌门、拟杆菌门和放线菌门为主。在属水平上,链球菌属、副普雷沃菌属、黄曲霉属、小杆菌属、梭状芽孢杆菌属XI、Pelomonas和丁酸菌属的相对丰度在足月小样儿组中显著升高(P<0.05)。结论足月儿胎盘组织中可检测到细菌,足月小样儿与正常足月儿其母亲胎盘菌群的组成和丰度在属水平上存在差异。 Objective To explore the compositions and differences in maternal placental microbiota between full-term small-for-gestational-age(SGA)infants and normal full-term infants,and provide reference data for further research.Methods A total of 9 samples of placental tissues of pregnant women who were regularly examined and delivered in our hospital from January to September,2018 were collected and divided into full-term SGA infant group and normal full-term(NFT)infant group according to birth weight.The DNA of placental tissue microbiota was extracted by using a tissue DNA extraction kit;16S rDNA high-throughput sequencing technology was used to analyze the microflora in the samples.Results There was no difference in placental microbiota between the two groups at the phylum level(P>0.05),and both groups were mainly composed of Firmicutes,Proteobacteria,Bacteroides and Actinomycetes,while the relative abundances of Streptococcus,Paraprevotella,Flavonifractor,Dialister,ClostridiumⅪ,Pelomonas and Butyricicoccus in the SGA group were significantly higher than those in the NFT group at the genus level(P<0.05).Conclusion Bacteria can be detected in the placental tissue of full-term infants,and the composition and abundance of microflora are different between full-term SGA infants and normal full-term infants.
