栀子花提取物的制备及其糖代谢调节作用 被引量:1

摘要 为开发栀子花的营养与药用价值,采用单因素法和均匀设计法分析由超声波法获得的栀子花提取物,通过测定各提取物中环烯醚萜苷、黄酮、三萜的含量及各提取物对α-葡萄糖苷酶活性的抑制率来优化栀子花的提取工艺,并采用胰岛素抵抗的人肝癌HepG2细胞模型评价采用优化工艺获得的提取物对糖代谢的调节作用。结果显示,优化后的栀子花提取工艺为:乙醇体积分数80%、液料比25∶1(mL∶g)、时间120 min、温度60℃。此优化工艺对环烯醚萜苷、黄酮和三萜的提取率分别为2.61%、5.37%、2.84%,对α-葡萄糖苷酶活性的抑制率可达78.2%,能有效提高胰岛素抵抗细胞HepG2的糖代谢能力,降低细胞氧化应激和炎症反应,从而改善细胞胰岛素抵抗状态。 In order to develop the nutritional and medicinal value of Gardenia jasminoides Ellis flowers,the ultrasonic extraction of G.jasminoides flower extract was optimized by single factor method and uniform design method,and the indexes concerned included the contents of total iridoid glycoside,flavonoid and triterpene of different extracts,and their inhibitory effect onα⁃glycosidase.Last⁃ly,the effect of G.jasminoides extracts prepared under the optimized extraction condition on glycometabolism was evaluated using in⁃sulin⁃resistant HepG2 cell model.The results showed that the optimized extraction process of G.jasminoides flower was to use 80%(v/v)ethanol solution at liquid⁃material ratio of 25∶1(mL∶g)for 120 min at 60℃,with the contents of total iridoid glycoside,flavonoid and triterpene reaching 2.61%,5.37%and 2.84%,respectively.G.jasminoides flower extract prepared under the optimized condition could depress the activity ofα⁃glycosidase with an inhibitory rate of 78.2%,resulting in promoted glucose metabolism of insulin⁃resistant HepG2 cell,and reduced oxidative stress and inflammatory reaction,thereby alleviating the insulin resistance of HepG2 cells.
