SMC3 mRNA表达水平与免疫浸润协同对乳腺癌预后的影响

摘要 目的研究SMC3 mRNA表达水平与乳腺癌临床病理指标的相关性,探讨其表达水平与免疫细胞浸润、免疫检查点基因之间的关系及SMC3表达与免疫浸润协同对乳腺癌预后的影响。方法基于TCGA公共数据库分析SMC3与临床特征变量、免疫检查基因之间的相关性,应用TIMER2.0数据库分析SMC3 mRNA表达和突变与免疫细胞浸润之间的关系及SMC3 mRNA表达水平和免疫细胞浸润与临床预后的关系。结果SMC3 mRNA高表达与淋巴结转移多和TNM分期升高有关,SMC3 mRNA表达与B细胞、CD4+T细胞、CD8+T细胞、嗜中性粒细胞、巨噬细胞和髓样树突状细胞的免疫浸润呈正相关。在Luminal A型乳腺癌中,相比于野生型,SMC3突变型CD8+T细胞、嗜中性粒细胞和巨噬细胞具有较高的浸润水平。免疫检查点基因表达与SMC3 mRNA表达呈正相关。SMC3 mRNA表达与免疫浸润水平协同影响乳腺癌的预后。结论SMC3 mRNA表达水平与乳腺癌免疫细胞浸润相关,SMC3表达与免疫浸润协同作用于乳腺癌预后。 Objective To investigate the correlation between the expression of SMC3 mRNA and clinical characteristic,immune cell infiltration,immune checkpoint gene expression in breast cancer tissues,and to further explore the expression of SMC3 mRNA and the immune cell infiltration combined effect on the prognosis of breast cancer.Methods Based on the TCGA public database,we analyzed the association between SMC3 mRNA and clinical characteristic,immune checkpoint gene expression.Using TIMER2.0 database,we analyzed the association between SMC3 mRNA expression,mutation and immune cell infiltration and combined effect on the prognosis of breast cancer.Results The overexpression of SMC3 mRNA was positive correlated with lymph node metastasis and TNM staging.The expression level of SMC3 mRNA was positively correlated with B cell,CD8+T cells,Neutrophils,Macrophages and Myeloid dendritic cells infiltration.In Luminal A breast cancer,CD8+T cells,Neutrophils,and Macrophages were higher in the SMC3 mutant than in the wild SMC3 type.The expression of immune checkpoint gene was positively correlated with the expression of SMC3 mRNA.The expression of SMC3 and the level of immune infiltration affect the prognosis of breast cancer.Conclusion The expression of SMC3 mRNA is related to immune cell infiltration in the microenvironment of breast cancer,and they play a role in the prognosis of breast cancer.
