通络玉液汤对糖尿病合并心肌病小鼠心肌细胞凋亡的改善作用及相关信号通路的影响 被引量:1

Effect of Tongluo Yuye Decoction on apoptosis of myocardial cells and related signaling pathways in diabetic mice with cardiomyopathy

摘要 目的探讨通络玉液汤对糖尿病合并心肌病小鼠心肌细胞凋亡的改善作用及可能的作用机制。方法将10只SPF级db/m小鼠设为正常组,不予处理。将30只db/db小鼠,根据处理方式的不同,随机分为模型组、苯那普利组(Ben组)及通络玉液汤组(TY组)。从空腹血糖、心肌组织HE染色、左心室功能及相关蛋白表达水平等方面分析各组小鼠相关指标的差异。结果在2~8周,Ben组和TY组小鼠的空腹血糖明显降低(P<0.05)。模型组小鼠心肌细胞染色变浅,部分细胞出现凋亡,染色质深染,细胞核消失。Ben组及TY组小鼠心肌细胞凋亡细胞数量及渗出均减少,结构相对整齐。Ben组和TY组的LVISD、EF、FS、LVMI、HWI值明显升高(P<0.05)。Ben组和TY组小鼠心肌组织中Bax蛋白、Caspase-3蛋白表达水平明显降低,Bcl-2蛋白表达水平升高。结论通络玉液汤可能通过调控凋亡相关蛋白表达水平,改善糖尿病小鼠心肌组织的损伤程度。 Objective To explore the improvement effect of Tongluo Yuye Decoction on cardiomyocyte apoptosis in diabetic mice with cardiomyopathy and its possible mechanism.Methods Ten SPF db/m mice were as normal group.Thirty db/db mice were divided into three groups:model group,Ben group and TY group.The differences of related indicators of each group mice were analyzed from the aspects of fasting blood glucose,myocardial tissue HE staining,left ventricular function and related protein expression levels.Results In 2~8 weeks,fasting blood glucose was significantly reduced in the Ben group and TY group(P<0.05).The myocardial cells in the model group were lighter stained,some apoptotic cells appeared,and the chromatin was deep stained.The number and exudation of cardiomyocyte apoptotic cells were less in Ben group and TY group.The LVISD,EF,FS,LVMI and HWI values of Ben group and TY group were significantly higher(P<0.05).The expressions of Bax and the Caspase-3 in Ben and TY groups were decreased,while the Bcl-2 was increased.Conclusion Tongluo Yuye Decoction might improve the myocardial injury of diabetic mice by regulating the expression of apoptosis-related proteins.
