绞股蓝多糖对热应激小鼠睾丸氧化损伤的保护作用

Protective effect of Gynostemma pentaphyllum polysaccharide on oxidative damage to testis in mice under heat stress

摘要 为了探究不同剂量绞股蓝多糖(Gynostemma pentaphyllum polysaccharides,GPP)对热应激引起的小鼠睾丸氧化损伤的保护作用,本研究采用生物化学、酶联免疫吸附、免疫组化等技术分别对空白对照(Con),热应激对照(HS),绞股蓝多糖高(GPP-H)、中(GPP-M)、低剂量(GPP-L)组小鼠的睾丸指数、睾丸细胞凋亡、雄激素受体表达以及生精小管直径、睾丸组织抗氧化能力进行观察和测定分析。结果发现,GPP-L组和GPP-M组血清睾酮含量与HS组相比均显著升高(P<0.05);GPP-M组和GPP-L组的丙二醛(MDA)含量与HS组相比显著降低(P<0.05),谷胱甘肽过氧化物酶(GSH-Px)含量、过氧化氢酶(CAT)活力、超氧化物歧化酶(SOD)活力与HS组相比显著升高(P<0.05);GPP各剂量组睾丸曲精小管直径与HS组相比均显著增大(P<0.05);各GPP剂量组凋亡细胞累计光密度值/视野下睾丸组织的面积(IOD/Area)较HS组均有所减小,其中,GPP-M组显著减小(P<0.05);各GPP剂量组AR阳性细胞IOD/Area较HS组均有所增大,其中GPP-L和GPP-M组雄性激素受体(AR)表达程度显著增强(P<0.05)。结果表明,GPP可以改善热应激导致的睾丸组织损伤和细胞凋亡,保护AR蛋白的正常表达以及提高热应激小鼠睾丸抗氧化能力,其中GPP-M效果较好。 In order to explore the protective effect of different doses of Gynostemma pentaphyllum polysaccharides(GPP)on oxidative dam⁃age to mouse testis caused by heat stress,biochemical,enzyme-linked immunosorbent assay and immunohistochemical techniques were used to observe and analyze the testicular index,testicular apoptosis,androgen receptor expression,seminiferous tubule diameter and testicular antioxidant capacity of mice.The results showed that the content of serum testosterone in the GPP-L group and the GPP-M group was signifi⁃cantly higher than that in the HS group(P<0.05),that the content of MDA in the GPP-M group and the GPP-L group was significantly lower than that in the HS group(P<0.05),that the content of GSH-Px,CAT activity and SOD activity were significantly higher than those in the HS group(P<0.05),and that the diameter of testicular seminiferous tubule in all the GPP groups was significantly larger than that in the HS group(P<0.05).The IOD/Area of apoptotic cells in each GPP group was significantly lower than that in the HS group(P<0.05),and the IOD/Area of AR positive cells in each GPP group was significantly higher than that in the heat stress group,and the expression of AR in the GPP-L and GPP-M groups was significantly increased(P<0.05).In conclusion,GPP alleviated testicular tissue injury and ap⁃optosis induced by heat stress,protected the normal expression of AR protein and enhanced the antioxidant capacity of testis in heat stressed mice,especially GPP-M mice.
