2-脱氧-D-葡萄糖抗肿瘤机制的研究进展

摘要 2-脱氧-D-葡萄糖(2-DG)作为一种葡萄糖类似物,可通过抑制肿瘤细胞有氧糖酵解、减少肿瘤细胞的能量生成等抑制肿瘤细胞增殖、侵袭与迁移等。近年来,随着2-DG药理机制研究的深入,其在分子层面的抗肿瘤作用机制也逐渐被人们揭示,包括影响蛋白质糖基化、调节信号通路以及阻滞细胞周期等。此外,2-DG也可协同其他治疗手段发挥抗肿瘤作用,提高肿瘤细胞对放化疗的敏感性。但目前关于2-DG的研究仅限于基础研究,其临床应用的安全性及有效性还有待未来进一步研究探讨。 As a glucose analogue,2-deoxy-D-glucose(2-DG)can inhibit the tumor cellproliferation,invasion and migration by inhibiting aerobic glycolysis and reducing the energy generation of tumor cells.In recent years,with the in-depth study on the pharmacological mechanism of 2-DG,its anti-tumor mechanism at the molecular level has been gradually revealed,including its influence on protein glycosylation,regulating of signaling pathways and cell cycle arrest.In addition,2-DG can cooperate with other therapies to exert anti-tumor effects,and increase the sensitivity of tumor cells to radiotherapy and chemotherapy.However,the current researches on 2-DG are limited to basic research,and the safety and effectiveness of its clinical application need to be further explored in the future.
