摘要
目的:探讨肝细胞癌(HCC)患者外周血T细胞表面T细胞免疫球蛋白粘蛋白-3(Tim-3)的表达,以及氩氦刀治疗对Tim-3表达的影响。方法:收集2019年7月至2021年1月于郑州大学第五附属医院进行治疗的50例HCC患者的外周静脉血标本,其中38例患者进行了肝脏肿块氩氦刀治疗,治疗后再次收集外周静脉血标本,同时采集26例健康志愿者外周静脉血标本。采用流式细胞术检测HCC患者及健康志愿者外周血T细胞Tim-3的表达,检测HCC患者外周血Tim-3阳性及Tim-3阴性T细胞早期凋亡分子[膜联蛋白(Annexin-V)抗体]和内因子[干扰素-γ(IFN-γ)]的表达水平,比较38例HCC患者行氩氦刀治疗前后外周血T细胞Tim-3的表达水平。结果:HCC患者外周血T细胞Tim-3的表达高于健康志愿者[(14.351±3.282)%比(5.564±1.750)%,P<0.01]。HCC患者Tim-3阳性T细胞分泌IFN-γ水平低于Tim-3阴性T细胞[(17.647±3.435)%比(25.439±4.151)%,P<0.01]。HCC患者Tim-3阳性T细胞Annexin V表达水平高于Tim-3阴性T细胞[(18.180±3.102)%比(9.479±2.371)%,P<0.01]。38例HCC患者经氩氦刀治疗前的外周血T细胞Tim-3的表达水平与治疗后比较差异未见统计学意义[(14.251±3.383)%比(13.556±2.795)%,P>0.05]。结论:HCC患者外周血T细胞Tim-3表达增高,导致T细胞功能耗竭,Tim-3可作为HCC的有效治疗靶点,氩氦刀治疗对HCC患者Tim-3表达无明显影响。
Objective To investigate the expression of T cell immunoglobulin and mucin-containing molecule(Tim-3)on the surface of T cells in peripheral blood in patients with hepatocellular carcinoma(HCC),and the effect of argon-helium knife therapy on Tim-3 expression.Methods Peripheral venous blood samples were collected from 50 HCC patients who were treated in the Fifth Affiliated Hospital of Zhengzhou University from July 2019 to January 2021.Among them,38 patients underwent argon-helium knife treatment for liver masses,after treatment,the peripheral blood samples were collected again;at the same time,peripheral venous blood samples were collected from 26 healthy volumteers.The expressions of Tim-3 in the selected HCC patients and healthy volumteers were determined by flow cytometry;expression levels of the early apoptotic molecules(Annexin V antibody)and endogenous factor interferon-γ(IFN-γ)in peripheral blood Tim-3 positive and Tim-3 negative T cells in HCC patients were detected;the Tim-3 expression level on peripheral blood T cells in 38 HCC patients before and after argon-helium knife treatment were compared.Results The expression of Tim-3 on peripheral blood T cells in HCC patients was higher than that in healthy volunteers,(14.351±3.282)%vs.(5.564±1.750)%,P<0.01.The level of IFN-γin Tim-3 positive T cells was lower than that in Tim-3 negative T cells in HCC patients,(17.647±3.435)%vs.(25.439±4.151)%,P<0.01.The expression level of Annexin V on Tim-3 positive T cells was higher than that on Tim-3 negative T cells in HCC patients,(18.180±3.102)%vs.(9.479±2.371)%,P<0.01.There was no statistically significant difference in preoperative and postoperative expression level of Tim-3 on peripheral blood T cells in the 38 HCC patients treated by argon-helium knife,(14.251±3.383)%vs.(13.556±2.795)%,P>0.05.Conclusions The expression of Tim-3 on peripheral blood T cells in HCC patients increases,leading to depletion of T cell function.Tim-3 can be an effective therapeutic target for HCC,but argon-helium knife therapy has no significant effect on Tim-3 expression in HCC patients.
