miR-506对结肠癌细胞增殖和转移活性的影响

摘要 目的:观察miR-506对结肠癌细胞活性、增殖、转移等恶性表型的影响。方法:以结肠癌细胞系HCT116为模型,以处理方式不同分为pcDNA3空载体对照组、过表达miR-506组、pSIH1空载体对照组及抑制miR-506组。荧光实时定量PCR检测各组中miR-506的表达水平,CCK8实验检测细胞的活性,集落形成实验检测细胞的集落形成能力,Transwell小室实验检测细胞的迁移和侵袭能力。结果:与pcDNA3空载体对照组比较,过表达miR-506后HCT116细胞内miR-506表达水平升高,差异有统计学意义(t=28.97,P<0.05),而细胞活性、集落形成能力、迁移和侵袭的能力减弱,差异有统计学意义(t=6.14、8.63、5.32、3.24,P<0.05);抑制miR-506组与pSIH1组比较,miR-506的相对表达量、细胞活性、集落形成能力、迁移和侵袭能力比较,差异无统计学意义(P>0.05)。结论:miR-506抑制结肠癌细胞HCT116活性、集落形成能力、侵袭和迁移能力,在结肠癌细胞中发挥抑癌基因的作用。 Objective:To observe the effect of miR-506 on the malignant phenotype of colon cancer cell activity,proliferation and metastasis.Methods:Colon cancer cell line HCT116 was used as a model and was divided into pcDNA3 empty vector control group,overexpression miR-506 group,pSIH1 empty vector control group and inhibition miR-506 group according to the treatment.Fluorescence real-time quantitative PCR was performed to detect the expression level of miR-506 in each group,and CCK8 assay was performed to detect the activity of cells.Colony formation assay was performed to detect the colony formation ability of cells,and Transwell assay was performed to detect the migration and invasion ability of cells.Results:The level of miR-506 expression in HCT116 cells was elevated after overexpression of miR-506 compared with pcDNA3 empty vector control,and the difference was statistically significant(t=28.97,P<0.05).Cell activity,colony formation capacity,migration and invasion were diminished,with statistically significant differences(t=6.14,8.63,5.32,3.24,P<0.05).The differences in the relative expression of miR-506,cell activity,colony formation ability,migration and invasion ability were not statistically significant in the miR-506 inhibition group compared with the pSIH1 group(P>0.05).Conclusion:miR-506 inhibits HCT116 activity,colony-forming ability,invasion and migration ability of colon cancer cells and functions as an oncogene in colon cancer cells.
