摘要
目的:建立并评价经改良翼点入路开颅构建食蟹猴缺血再灌注脑卒中模型。方法:取食蟹猴6只,分为假手术组3只,建模组3只。假手术组仅行手术,术中不阻断大脑中动脉;手术组术中夹闭大脑中动脉1h后松开。术毕,动物清醒且生命征平稳后方送回动物房。记录并比较两组食蟹猴术前及术后神经功能评分、定量运动活动量。结果:6只食蟹猴术后均存活;与术前相比,假手术组的神经功能无明显损伤、定量运动活动量无显著差异;建模组的神经功能损伤符合脑卒中表现、定量运动活动量较术前明显降低。结论:经改良翼点入路开颅构建食蟹猴大脑中动脉缺血再灌注脑卒中模型方法可行。
Objective To establish and evaluate the construction of an ischemic reperfusion stroke model in crab eating macaques through improved approach for pterional craniotomy.Methods Six crab-eating macaques were divided into three in the sham-surgery group and three in the model group.In the sham-surgery group,only operation was performed without blocking the middle cerebral artery;The model group clamped the middle cerebral artery for 1 hours before releasing it.After the surgery was completed,the animal was brought back to the animal room only after it was clear and had stable vital signs.Record and compare the preoperative and postoperative neurological function scores and quantitative motor activity between two groups of crab-eating macaques.Results The results showed that all six crab-eating macaques survived after surgery.Compared with that before operation,the nerve function of the sham-surgery group had no obvious damage and there was no significant difference in quantitative motor activity;The neural function damage in the model group was consistent with stroke symptoms,and the quantitative motor activity was significantly reduced compared to preoperative.Conclusions The method of constructing a middle cerebral artery ischemia-reperfusion stroke model in crab-eating macaques through pterional craniotomy is feasible.
