SMARCA4在恶性肿瘤中的研究进展

摘要 SWI/SNF相关基质关联肌动蛋白依赖染色质调控因子亚家族a成员4(SMARCA4)基因与恶性肿瘤的关系密切,人类恶性肿瘤中存在SMARCA4的突变、过表达和表观遗传沉默。SMARCA4在肿瘤中具有双重作用,SMARCA4可作为致癌基因促进肿瘤细胞的增殖、迁移、抗细胞凋亡等,影响肿瘤的发生和发展;SMARCA4还可作为抑癌基因在一些肿瘤中失活突变,且SMARCA4缺失型肿瘤的恶性程度很高,分化低,预后差。因此,阐明SMARCA4在肿瘤中的作用及其相关分子机制对肿瘤的诊断和治疗具有重要意义。 SWI/SNF related matrix associated actin dependent regulator of chromatin subfamily a member 4(SMARCA4)genes are closely associated with malignant tumors,and SMARCA4 mutations,overexpression,and epigenetic silencing are found in human malignant tumors.SMARCA4 plays a dual role in tumor:SMARCA4 can act as an oncogene to promote the proliferation,migration and anti-apoptosis of tumor cells,and affect the occurrence and development of tumors;SMARCA4 can also act as a tumor suppressor gene to inactivate mutations in some tumors,and SMARCA4 deletion tumors have a high degree of malignancy,low differentiation,and poor prognosis.Therefore,understanding the role of SMARCA4 in tumor and its related molecular mechanism is of great significance for the tumor diagnosis and treatment.
