数字PCR检测循环肿瘤非编码RNA在肺癌液体活检中的应用进展

摘要 数字聚合酶链反应(dPCR)作为近年来有重大技术突破并仍在发展的核酸定量分析技术,凭借其不依赖循环阈值、无需内参基因及标准曲线即可实现绝对定量的特点和灵敏度高、重现性好、酶抑制剂耐受性高、参与单细胞基因表达分析准确性好等优点,在分子生物学研究中具有广泛的应用前景。目前,有研究逐渐将其应用于肺癌患者的循环非编码RNA检测,发现dPCR凭借其优良检测效能在肺癌的液体活检领域中有较大应用潜力,可为肺癌液体活检的检测和研究提供新的技术手段和思路。 Digital polymerase chain reaction(dPCR),as a nucleic acid quantitative analysis technology that has made major technological breakthroughs in recent years and is still developing.It has the features of independence from cycle threshold value,no need for internal reference genes and standard curve to achieve absolute quantification,as well as the advantages of high sensitivity,good reproducibility,high enzyme inhibitor tolerance,and good accuracy in single-cell gene expression analysis,therefore has a wide range of application prospects in the field of molecular biology.At present,studies are gradually applying it to the detection of circulating non-coding RNA in lung cancer patients,and it is found that dPCR has great application potential in the field of liquid biopsy of lung cancer with its excellent detection efficiency,which can provide new technical means and ideas for the detection and research of lung cancer.
