H9N2亚型禽流感病毒感染鸡红细胞引起白细胞介素的免疫应答 被引量:1

Immune Responses of Interleukin Induced by H9N2 Subtype Avian Influenza Virus Infection in Chicken Erythrocytes

摘要 为研究H9N2亚型禽流感病毒(H9N2 AIV)感染鸡红细胞后白细胞介素(ILs)参与的免疫应答反应,进一步揭示鸡红细胞在感染H9N2亚型禽流感病毒中所发挥的免疫调节作用,应用实时荧光定量PCR技术分别对体外感染H9N2 AIV后0、2、6、10 h和体内攻毒3、7、14 d后8种白细胞介素相关基因(IL-6、IL-7、IL-12、IL-13、IL-15、IL-16、IL-18、IL-34)的表达水平进行检测。结果表明,8种ILs相关基因均在鸡红细胞中表达,其表达水平在体外感染H9N2 AIV后和体内攻毒后呈现动态变化;与对照组相比,体外感染H9N2 AIV后不同时间点IL-6和IL-18的相对表达量呈现逐渐升高的趋势,而IL-7、IL-12、IL-15的相对表达量呈现逐渐下降的趋势;IL-13在感染H9N2 AIV后的0、6 h显著下降,而IL-16仅在感染H9N2 AIV后的2 h显著下降。体内攻毒后的不同时间点,IL-12、IL-13、IL-15、IL-16的相对表达水平均逐渐升高,而IL-6、IL-18、IL-34的表达水平先升高后降低,仅IL-7表达水平先降低后升高。综上,鸡感染H9N2 AIV后红细胞发生了相应的免疫应答反应,并通过促炎因子以及抗炎因子的表达来调节鸡感染H9N2后的免疫应答。 To explore the immune responses of interleukin(ILs)during the infection in chicken erythrocytes induced by H9N2 subtype avian influenza virus(H9N2 AIV)and further reveal immunomodulatory role of chicken erythrocytes in H9N2 subtype avian influenza virus infection in chickens,in this study,real-time quantitative PCR was used to detect the expression levels of eight IL-related genes(IL-6,IL-7,IL-12,IL-13,IL-15,IL-16,IL-18,IL-34)in chicken erythrocytes infected by H9N2 AVI at 0,2,6,and 10 h,as well as their expression levels in erythrocytes of H9N2-infected chicken were detected at 3,7 d and 14 d of poison attack in vivo.The results showed that expression of eight IL-related genes in chicken erythrocytes was identified and their expression levels showed dynamic changes after infection and challenge.Compared with the control group,the relative expression levels of IL-6 and IL-18 showed a increasing trend,while the relative expression levels of IL-7,IL-12,and IL-15 showed a declining trend in vitro.In addition,the expression level of IL-13 significantly declined at 0 h and 6 h after infection,while the expression level of IL-16 only significantly declined at 2 h.At different time of in vivo,the relative expression levels of IL-12,IL-13,IL-15 and IL-16 increased gradually,while the expression levels of IL-6,IL-18 and IL-34 increased at first and then declined,only the expression level of IL-7 declined at first and then increased.The results above indicated that chicken erythrocytes participated in the immune responses after infection of H9N2 AVI,and the immune responses of chickens infected with H9N2 was regulated by expression of pro-inflammatory factors and anti-inflammatory factors.
