基于斑马鱼模型的玉叶金花三萜皂苷的肝保护作用研究

摘要 目的:研究玉叶金花三萜皂苷(YYTS)的肝保护作用。方法:采用对乙酰氨基酚(APAP,8 mm)诱导斑马鱼肝损伤模型,药浴后的斑马鱼幼鱼进行组织匀浆,测定ALT、AST含量。斑马鱼成鱼给药48 h后,观察YYTS对斑马鱼肝脏明亮度和卵黄囊吸收的影响;采用qPCR法检测斑马鱼IL-1β、IL-6 mRNA表达。结果:与模型组比较,0.11、0.33、1.00μg/mL组斑马鱼幼鱼肝组织ALT、AST水平明显降低(P<0.05),其中0.33、1.00μg/mL组作用明显。0.11、0.33、1.00μg/mL YYTS组斑马鱼肝变性改善作用分别为83.9%、91.3%、79.5%(P<0.05);0.33、1.00μg/mL YYTS组斑马鱼卵黄囊吸收延迟改善作用分别为32.6%、40.4%(P<0.05)。与模型组比较,0.33、1.00μg/mL YYTS组斑马鱼IL-1β、IL-6 mRNA的相对表达量显著降低(P<0.05)。结论:YYTS具有较好的肝保护作用。 Objective:To study the liver protective effect of triterpenoid saponin from Mussaenda pubescens(YYTS).Methods:A zebrafish liver injury model was induced by acetaminophen(APAP,8 mM).Tissue homogenization was performed on zebrafish juveniles after drug bath,and the levels of ALT and AST were measured.After 48 hours of administration to adult zebrafish,the effect of YYTS on the liver brightness and yolk sac absorption of zebrafish were observed.The expression of IL-1βand IL-6 mRNA in zebrafish was detected by qPCR.Results:Compared with the model group,the levels of ALT and AST in the liver tissue of juvenile zebrafish in the 0.11,0.33,and 1.00μg/mL groups were significantly reduced(P<0.05),with the 0.33 and 1.00μg/mL groups having a significant effect.The improvement effects of 0.11,0.33,and 1.00μg/mL YYTS on liver degeneration in zebrafish were 83.9%,91.3%,and 79.5%,respectively(P<0.05).The improvement effect of delayed absorption of zebrafish yolk sac in the 0.33 and 1.00μg/mL YYTS groups was 32.6%and 40.4%,respectively(P<0.05).Compared with the model group,the relative expression levels of IL-1βand IL-6 mRNA in zebrafish in 0.33 and 1.00μg/mL YYTS groups were significantly decreased(P<0.05).Conclusion:YYTS has a good liver protective effect.
