circRNA_MYLK在膀胱癌中的表达量及其临床价值

Expression of circRNA_MYLK in bladder cancer and its clinical value

目的探究环状RNA肌球蛋白轻链激酶(circRNA_MYLK)在膀胱癌中的表达量及其与患者临床特征和预后的关系。方法收集2015年1月至2017年6月行手术治疗的68例患者的膀胱癌组织和癌旁组织标本。采用实时荧光定量聚合酶链反应(qRT-PCR)检测circRNA_MYLK在膀胱癌组织和癌旁组织中的表达量;分析circRNA_MYLK与患者临床病理特征的关系;用Cox回归模型分析影响患者预后的因素。结果膀胱癌组织中circRNA_MYLK的表达量高于癌旁组织(P<0.01)。膀胱癌TNM分期T_(3)~T_(4)期组织中circRNA_MYLK的表达量高于TNM分期T_(1)~T_(2)期组织(P<0.05)。circRNA_MYLK低表达组和高表达组的年龄、性别、肿瘤大小无显著差异(P>0.05);circRNA_MYLK低表达组和高表达组的远处转移、TNM分期、肿瘤分化程度具有显著差异(P<0.05)。Kaplan-Meier生存曲线显示,circRNA_MYLK高表达组的生存率低于circRNA_MYLK低表达组(P=0.033)。单因素Cox回归分析显示,远处转移、TNM分期、肿瘤分化程度、circRNA_MYLK表达量与患者预后相关(P<0.05)。多因素Cox回归分析显示,远处转移、高TNM分期、低肿瘤分化程度、circRNA_MYLK高表达是影响膀胱癌患者预后的独立危险因素(P<0.05)。结论circRNA_MYLK的表达与膀胱癌患者预后呈负相关,是预测预后的独立因素。

Objective To explore the expression of circRNA myosin light chain kinase(circRNA_MYLK)in bladder cancer and its relationship with clinical characteristics and prognosis of patients.Methods Bladder cancer tissues and adjacent tissues of 68 patients who underwent surgical treatment from January 2015 to June 2017 were collected.The expression of circRNA_MYLK in bladder cancer tissues and adjacent tissues was detected by quantitative real-time polymerase chain reaction(qRT-PCR);the relationship between circRNA_MYLK and clinicopathological features of patients was analyzed;the factors affecting the prognosis of patients was analyzed by Cox regression model.Results The expression of circRNA_MYLK in bladder cancer tissues was higher than that in adjacent tissues(P<0.01).The expression of circRNA_MYLK in bladder cancer tissues with TNM stage T_(3)-T_(4) was higher than that in tissues with TNM stage T_(1)-T_(2)(P<0.05).There were no significant differences in age,gender and tumor size between the circRNA_MYLK low expression group and the high expression group(P>0.05);there were statistically significant differences in distant metastasis,TNM stage and tumor differentiation between the circRNA_MYLK low expression group and the high expression group(P<0.05).Kaplan-Meier survival curve showed that the survival rate of circRNA_MYLK high expression group was lower than that of the circRNA_MYLK low expression group(P=0.033).Univariate Cox regression analysis showed that distant metastasis,TNM stage,tumor differentiation and circRNA_MYLK expression were related to the prognosis of patients(P<0.05).Multivariate Cox regression analysis showed that distant metastasis,high TNM stage,low tumor differentiation and high expression of circRNA_MYLK were independent risk factors affecting the prognosis of patients with bladder cancer(P<0.05).Conclusion The expression of circRNA_MYLK is negatively correlated with the prognosis of patients with bladder cancer and is an independent factor for predicting prognosis.
