阻燃剂与农药联合暴露致甲状腺癌的初步分析 被引量:1

摘要 目的 从基因水平探讨阻燃剂与农药联合暴露致甲状腺癌可能的生物联合作用机制。方法 通过比较毒理基因组学数据库(comparative toxicogenomics database,CTD)中获取阻燃剂与农药有确切证据的甲状腺癌相关基因,对阻燃剂与农药共同基因进行基因富集分析,获取共同基因/蛋白可能参与的生物进程或信号通路。结果 经分析共同作用关键基因61个,这些基因分别参与激素代谢过程、对有毒物质的反应、对无机物的反应、细胞对脂质的反应、细胞对激素刺激的反应等GO生物学进程和AMPK信号通路。结论 阻燃剂和农药主要通过肝微粒体酶P450酶系相关基因联合作用影响甲状腺激素水平,引起机体内分泌紊乱造成甲状腺功能改变,从而导致甲状腺癌的发生. Objective To explore the possible biological mechanisms of combined exposure of flame retardants and pesticides to thyroid cancer at the genetic level.Methods Thyroid cancer related genes with clear evidence of flame retardants and pesticides were obtained from the comparative toxicogenomics database(CTD).Gene enrichment analysis was performed on the common genes of flame retardants and pesticides to obtain the biological processes or signaling pathways that the common genes/proteins may be involved in.Results The results showed that 61 key genes were involved in the process of hormone metabolism,response to toxic substances,response to inorganic substances,cell response to lipids,cell response to hormone stimulation,and other GO biological processes and AMPK signaling pathways.Conclusion Flame retardants and pesticides mainly affect thyroid hormone levels through the combined action of liver microsomal enzyme P450 related genes,causing endocrine disorders and thyroid function changes,leading to the occurrence of thyroid cancer.
