摘要
目的探究肿瘤坏死因子-α诱导蛋白8-1(TNFAIP8L1,TIPE1)在结肠癌及癌旁正常组织的表达,及其表达水平与结肠癌患者临床病理参数和预后生存的关系。方法收集青岛大学附属医院2013年1月至2014年12月间诊断为结肠癌的患者的癌和癌旁正常组织。采用免疫组织化学(IHC)染色法检测样本中TIPE1的表达以分析其与患者年龄、性别、肿瘤大小、TNM分期、淋巴结转移、远处转移、病理分期等的相关性。组间比较采用t检验,患者预后生存采用Kaplan-Meier生存分析及单因素/多因素COX回归分析。结果TIPE1在结肠癌组织中的表达低于正常结肠组织(57.2±31.1比86.9±35.6,t=8.304,P<0.01)。共纳入181例结肠癌患者,TIPE1高表达患者69例,低表达112例。TIPE1高表达组患者平均生存期时间高于低表达组患者(80.9个月比59.0个月,Log-rank=14.615,P<0.01)。同时,COX回归分析提示,TIPE1表达水平为影响结肠癌患者预后的独立因素之一[风险比(HR)=0.538,95%可信区间(CI):0.307~0.942,P<0.05]。此外,TIPE1的表达水平与结肠癌患者的T分期(χ^(2)=7.578,P<0.05)、N分期(χ^(2)=17.899,P<0.01)、TNM分期(χ^(2)=16.772,P<0.01)、肿瘤位置(χ^(2)=14.832,P<0.01)、淋巴结转移与否(χ^(2)=14.119,P<0.01)以及肿瘤分化程度(χ^(2)=5.766,P<0.05)存在相关性。结论结肠癌中TIPE1的表达水平低于正常结肠组织,且与肿瘤进展及患者预后显著相关。
Objective To investigate the expression of tumor necrosis factor-α-induced protein 8 like 1(TNFAIP8L1,TIPE1)in colon cancer and adjacent normal tissues,and the correlation between TIPE1 and clinicopathological data or prognostic survival of patients with colon cancer.Methods Tumor tissues and normal tissues adjacent to the cancer were collected from patients diagnosed with colon cancer between January 2013 and December 2014 at the Affiliated Hospital of Qingdao University.Immunohistochemical(IHC)staining was used to detect the expression of TIPE1 in the samples and to analyze its correlation with age,sex,tumor size,TNM stage,lymph node metastasis,distant metastasis,pathological stage.The t-test was used for comparison between groups,theχ^(2) test was used for correlation analysis,and Kaplan-Meier survival analysis and one-way or multi-factor COX regression analysis were used for prognosis and survival of patients.Results IHC staining results showed that the TIPE1 expression was lower in colon cancer tissue than in normal colon tissue(57.2±31.1 vs.86.9±35.6,t=8.304,P<0.01).A total of 181 patients were included,including 69 patients with high TIPE1 expression and 112 patients with low TIPE1 expression.The mean survival time of patients with high TIPE1 expression was higher than that of patients with low TIPE1 expression(80.9 months vs.59.0 months,Log-rank=14.615,P<0.01).Meanwhile,COX regression analysis suggested that TIPE1 expression level was one of the independent prognostic factors of colon cancer patients[hazard ratio(HR=0.538,95%confidence interval(CI):0.307-0.942,P<0.05].In addition,the expression level of TIPE1 was positively correlated with T-stage(χ^(2)=7.578,P<0.05),N-stage(χ^(2)=17.899,P<0.01),TNM-stage(χ^(2)=16.772,P<0.01),tumor location(χ^(2)=14.832,P<0.01),and lymph node metastasis in patients with colon cancer(χ^(2)=14.119,P<0.01)and the degree of tumor differentiation(χ^(2)=5.766,P<0.05).Conclusion The expression of TIPE1 in colon cancer was lower than in normal colon tissue and was significantly associated with tumor progression and patient prognosis.
