摘要
为获得大孔树脂纯化光甘草定的最佳条件,以野生光果甘草的根及根茎为原料,通过考察21种不同类型大孔树脂对光甘草定的吸附率和解吸率的大小,筛选出一种适合纯化光甘草定的树脂。在此基础上,选择对纯化工艺影响较大的5种因素,洗脱剂浓度、上样速度、上样浓度、洗脱流速、洗脱体积进行最佳条件的确定。结果表明,D101型大孔树脂对光甘草定的纯化效果最佳,其最佳洗脱剂的浓度为90%乙醇,上样速度0.4mL/min,上样量为每毫升大孔树脂上样2.05mg,洗脱流速0.27mL/min洗脱体积4BV,该条件下,光甘草定的纯化度可达到32.9%。研究为光甘草定的纯化工艺提供了新的技术路线,也为光果甘草提取物的研究和光甘草定应用提供参考。
In order to optimize the process of glabridin by macroporous resin from glycyrrhiza glabra aerial parts,moisture content,adsorption rate and desorption rate of 21 resins were investigated,and the resin with the best effect was selected.Five factors(ethanol concentration,sample loading speed,sample con-centration,elution flow rate and elution volume)that have a great influence on the purification process were selected.The results showed that:D101 macroporous resin had the best purification effect on glabrid-in,the best eluent concentration was 90%ethanol,the eluent concentration was 0.4 mL/min,the sample concentration was 2.05mg of macroporous resin per ml,the elution flow rate was 0.27 mL/min,and the e-lution volume was 4BV.Under these conditions,the purification degree of glabridin could reach 32.9%.This study provides a new technical route for the purification of glycyrrhiza glabra,and also provides a ref-erence for the research of glycyrrhiza glabra extract and the application of glabridin.
