摘要
目的研究北苍术Atractylodes chinensis(DC.)Koidz.的化学成分及其血管新生抑制活性。方法北苍术95%乙醇提取物采用大孔树脂、硅胶、Sephadex LH-20、TLC、半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用MTT法测定其对人脐静脉内皮细胞HUVEC的细胞增殖抑制活性。结果从中分离得到11个化合物,分别鉴定为seco-愈创木酮(1)、gibberodione(2)、junipercamphor(3)、2-one-3,4-dehydro-4,15-dihydrocostic ester(4)、7α-hydroxycostol(5)、elema-1,3,11(13)-trien-12-oic acid(6)、atractylenolid I(7)、atractylenolidⅢ(8)、atrchiterpene B(9)、olean-12-ene-3,11-dione(10)、δ-amyrione(11)。化合物1~2、5对HUVEC细胞增殖的抑制率分别为(50.5±1.1)%、(47.3±0.9)%、(45.2±1.3)%。结论化合物1为新化合物,化合物2、4~6、10~11为首次从该植物中分离得到。化合物1~2、5具有一定程度的HUVEC细胞增殖抑制活性。
AIM To study the chemical constituents from Atractylodes chinensis(DC.)Koidz.and their angiogenesis inhibitory activities.METHODS The 95%ethanol extract from A.chinensis was isolated and purified by macroporous resin,silica gel,Sephadex LH-20,TLC and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their cell proliferation inhibitory activities on human umbilical vein endothelial cells HUVEC were measured by MTT assay.RESULTS Eleven compounds were isolated and identified as seco-guaione(1),gibberodione(2),junipercamphor(3),2-one-3,4-dehydro-4,15-dihydrocostic ester(4),7α-hydroxycostol(5),elema-1,3,11(13)-trien-12-oic acid(6),atractylenolid I(7),atractylenolidⅢ(8),atrchiterpene B(9),olean-12-ene-3,11-dione(10),δ-amyrione(11).The inhibitory rates of compounds 1-2,5 on the proliferation of HUVEC cells were(50.5±1.1)%,(47.3±0.9)%,and(45.2±1.3)%,respectively.CONCLUSION Compound 1 is a new compound.Compounds 2,4-6,10-11 are isolated from this plant for the first time.Compounds 1-2,5 have certain angiogenesis inhibitory activities in HUVEC cells.
