甘蔗与斑茅杂交后代的SSR标记鉴定方法

摘要 为简化甘蔗与斑茅杂交后代鉴定程序、缩短鉴定时间,提高检测准确性,本研究在前期简化基因组调研测序开发的SSR引物基础上,选取部分引物对其在甘蔗DNA混液、斑茅DNA混液、主要栽培种及甘蔗与斑茅杂交后代DNA的扩增情况进行分析。结果表明,选取的381对引物在斑茅DNA混液和10个甘蔗栽培种DNA混液中有363对引物在斑茅混液中检测到扩增产物,其中86对只在斑茅混液检测到扩增产物。将上述86对引物在192个甘蔗栽培种中进行扩增分析,筛选得到4对仅在斑茅及甘蔗与斑茅的后代有特异扩增条带的特异性引物。本研究可实现简单、快速、准确鉴定甘蔗和斑茅杂交后代的斑茅血缘,有利于高效地获得真实的杂种后代,加快甘蔗和斑茅杂交后代育种进程。 In order to simplify the identification procedure of the hybrid offspring of Saccharum and Erianthus arundinaceus,shorten the identification time and improve the detection accuracy.In this study,based on the SSR primers developed for simplified Erianthus arundinaceus genome sequencing,some primers were selected to analyze the DNA amplification conditions of Saccharum DNA mixture,Erianthus arundinaceus DNA mixture,hybrid progeny of Saccharum and Erianthus arundinaceus,and main cultivars.The results showed that among the selected 381 pairs of primers in the mixture of Erianthus arundinaceus DNA and 10 Saccharum cultivar DNA,363 pairs of primers detected amplification products in the Erianthus arundinaceus mixture,of which 86 pairs only detected amplification products in the Erianthus arundinaceus mixture.The above 86 pairs of primers were amplified and analyzed in 192 Saccharum cultivars,and 4 pairs of specific primers were screened to have specific amplification bands only in Erianthus arundinaceus and the offspring of Saccharum and Erianthus arundinaceus.This study can realize the simple,rapid and accurate identification of the Erianthus arundinaceus blood relationship of the hybrid offspring of Saccharum and the Erianthus arundinaceus,which is beneficial to obtain the true hybrid offspring efficiently and speed up the breeding process of the hybrid offspring of Saccharum and Erianthus arundinaceus.
