火石花多毛外植体初代培养成活率的分析

摘要 前期研究发现,火石花整株多毛,各类外植体的初代培养成活率均在5%左右,不利于火石花的离体快繁。为此,本研究采用2.0 mg/L的6-BA浇灌盆栽火石花,15 d后将获得的无毛嫩芽作为外植体进行初代培养,其成活率超过了61.11%。此后,无菌苗在含有6-BA 2.0 mg/L和NAA 0.1 mg/L的MS培养基中增殖(增殖系数高达10.37),在含有NAA 0.1 mg/L的MS培养基中生根(生根率100%),生根苗的叶背纤维发育良好。经炼苗后,实现了火石花组培苗的成功移栽(成活率98.65%以上),移栽苗开花正常。本研究显著提升了火石花多毛外植体初代培养的成活率,建立了火石花优质种苗的离体快繁体系,为其他多毛植物的初代培养提供了新的参考。 Previous stu die s found that the whole plant of O.delavayi is hairy,and the survival rate of various explants in primary culture is about 5%,which is not conducive to the rapid propagation of O.delavayi in vitro.In this study,we used 2.0 mg/L 6-BA to irrigate potted O.delavayi.After 15 days,the obtained hairless shoots were used as explants for primary culture,and the survival rate was more than 61.11%.After that,the aseptic plantlets proliferated in MS medium containing 2.0 mg/L 6-BA and 0.1 mg/L NAA(the proliferation coefficient was as high as 10.37),and rooted in MS medium containing 0.1 mg/L NAA(rooting rate 100%).The fibers on the back of leaves of rooting seedlings developed well.The results showed that the O.delavayi were transplanted successfully(the survival rate was above 98.65%),and the transplanted seedlings flowered normally.This study significantly improved the survival rate of the primary culture of the hairy explants of O.delavayi,and established the in vitro rapid propagation system of high-quality seedlings of O.delavayi,and provided a new reference for the primary culture of other hairy plants.
