伪原薯蓣皂苷联合运动对高脂血症大鼠肝脏脂代谢和抗氧化功能的影响

Effects of PPD Combined with Exercise on Liver Lipid Metabolism andAntioxidant Function in Hyperlipidemia Rats

摘要 伪原薯蓣皂苷(pseudoprotodioscin, PPD)是盾叶薯蓣(Dioscorea zingiberensis)中的一种甾体皂苷,可抑制胆固醇和甘油三酯合成。本研究考察了伪原薯蓣皂苷联合运动对高脂血症(HLP)大鼠肝脏脂代谢和抗氧化功能的影响。将大鼠分为C组、HLP组、E组、L-PPD组、M-PPD组、H-PPD组、E+H-PPD组,每组12只大鼠。C组为对照大鼠,其他组为高脂血症模型大鼠。C组和HLP组大鼠安静饲养。E组大鼠进行跑台运动。L-PPD组、M-PPD组、H-PPD组大鼠分别灌胃25、50、100 mg·kg-1·d-1的伪原薯蓣皂苷。E+H-PPD组大鼠在跑台运动前1 h灌胃100 mg·kg-1·d-1的伪原薯蓣皂苷溶液。干预时间为8周。分别检测各组大鼠的体重、血清脂代谢标志物(TC, TG, LDL-c, HDL-c, FFA)、肝脏抗氧化标志物(SOD, CAT和MDA)水平。通过HE染色观察大鼠肝脏形态。通过RT-qPCR检测肝脏SREBP1c、FAS、ABCA1、HMGCR、CYP7A1和ACC m RNA水平。通过Western blot检测肝脏Nrf2和Keap-1蛋白表达水平。结果显示,与E组和H+PPD组比较,E+H-PPD组大鼠的体重降低,血清TC、TG、LDL-c和FFA水平降低,血清HDL-c水平升高,肝脏空泡化脂滴减少,肝脏SOD和CAT水平升高,肝脏MDA水平降低,肝脏SREBP1c、FAS、HMGCR和ACC m RNA水平降低,肝脏ABCA1和CYP7A1 mRNA水平升高,肝脏细胞核Nrf2蛋白表达水平升高,肝脏Keap-1蛋白表达水平降低(P<0.05)。本研究表明,伪原薯蓣皂苷联合运动可纠正高脂血症大鼠肝脏脂代谢紊乱并提高抗氧化功能。 Pseudoprotodioscin(PPD)is a steroid saponin in Dioscorea zingiberensis,which inhibits the synthesis of cholesterol and triglycerides.This study investigated the effects of pseudoprotodioscin combined with exercise on liver lipid metabolism and antioxidant function in hyperlipidemia(HLP)rats.Rats were divided into C group,HLP group,E group,L-PPD group,M-PPD group,H-PPD group and E+H-PPD group,with 12 rats in each group.C group was the control rats,and other groups were hyperlipidemia model rats.Rats in C group and HLP group were fed quietly.Rats in E group did treadmill exercise.Rats in L-PPD group,M-PPD group and H-PPD group were intragastric with 25,50 and 100 mg·kg-1·d-1 pseudoprotodioscin,respectively.Rats in E+H-PPD group were given 100 mg·kg-1·d-1 pseudoprotodioscin solution 1 h before treadmill exercise.The duration of intervention was 8 weeks.Body weight,serum lipid metabolism markers(TC,TG,LDL-c,HDL-c,FFA)and liver antioxidant markers(SOD,CAT and MDA)were detected in each group.The liver morphology of rats was observed by HE staining.Liver SREBP1c,FAS,ABCA1,HMGCR,CYP7A1 and ACC mRNA levels were detected by RT-qPCR.The expression levels of Nrf2 and Keap-1 proteins in liver were detected by Western blot.The results showed that compared with E group and H+PPD group,the body weight of rats in E+H-PPD group decreased,the levels of TC,TG,LDL-c and FFA in serum decreased,the levels of HDL-c in serum increased,the vacuolated lipid droplets in liver decreased,the levels of SOD and CAT in liver increased,the levels of MDA in liver decreased,liver SREBP1c,FAS,HMGCR and ACC mRNA levels decreased,liver ABCA1 and CYP7A1 mRNA levels increased,liver nuclear Nrf2 protein expression level increased,liver Keap-1 protein expression level decreased(P<0.05).This study showed that pseudoprotodioscin combined with exercise could correct liver lipid metabolism disorder and improve antioxidant function in hyperlipidemia rats.
