摘要
目的:建立一测多评法(QAMS)同时测定菊花配方颗粒中绿原酸、1,3-O-二咖啡酰基奎宁酸、芦丁、木犀草苷、木犀草素-7-O-β-D-葡萄糖醛酸苷、3,4-O-二咖啡酰基奎宁酸、3,5-O-二咖啡酰基奎宁酸、4,5-O-二咖啡酰基奎宁酸的含量。方法:采用高效液相色谱法进行含量测定,色谱柱为Agilent TC-C_(18) (250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸水溶液为流动相梯度洗脱,检测波长为348 nm,流速为1.0 mL·min^(-1),柱温为30℃,进样量为10μL。以木犀草苷为内标物,采用多点校正法计算其他7个成分的相对校正因子,测定8个成分的含量,并比较与外标法测定结果的差异。结果:8个成分在一定质量浓度范围内与峰面积呈良好的线性关系,平均加样回收率为96.7%~102.1%,RSD为1.8%~2.9%。QAMS与外标法测定7批菊花配方颗粒中8个成分含量差异无统计学意义。结论:建立的QAMS简便快速,可以更全面地控制菊花配方颗粒的质量。
Objective:To establish a quantitative analysis of multiple components with a single marker(QAMS)method for simultaneous determination of the content of chlorogenic acid,1,3-O-dicaffeoylquinic,rutin,luteoloside,luteolin-7-O-β-D-glucuronide,3,4-O-dicaffeoylquinic acid,3,5-O-dicaffeoylquinic acid,4,5-O-dicaffeoylquinic acid in Juhua Formula Granules.Methods:High-performance liquid chromatography(HPLC)was used for content determination.The chromatographic column was Agilent TC-C18(250 mm×4.6 mm,5μm),and the mobile phase consisted of acetonitrile and 0.1%phosphoric acid aqueous solution in gradient elution.The detection wavelength was set at 348 nm,the flow rate was 1.0 mL·min^(-1),the column temperature was 30°C,and the injection volume was 10μL.Luteoloside was used as an internal standard,and the relative correction factors of the other seven components were calculated using a multi-point calibration method.The content of the eight components was determined,and the differences between the results obtained by QAMS and the external standard method were compared.Results:The eight components showed good linearity within a certain range,with average recovery rates ranging from 96.7%to 102.1%,and RSDs ranging from 1.8%to 2.9%.There was no statistically significant difference in the content of the eight components between QAMS and the external standard method in seven batches of Chrysanthemum formula granules.Conclusion:The established QAMS method is simple and rapid,and it can comprehensively control the quality of Juhua Formula Granules.
