摘要
通过单因素试验和正交试验对凝结芽孢杆菌Lys 274发酵条件和培养基进行优化,提高Lys 274的生物量和芽孢数。结果表明,最佳培养基组成为玉米淀粉10 g/L、葡萄糖2 g/L、乳糖5 g/L、酵母膏20 g/L、蛋白胨15 g/L、牛肉膏5 g/L、磷酸二氢钾4 g/L,硫酸锰0.2 g/L、氯化亚铁0.4 g/L,培养条件为装液量50 mL、pH 6.0、37℃条件下振荡培养44 h。其生物量达到5.60×10~9 CFU/mL,芽孢数达到4.47×10~9 CFU/mL,芽孢率达79.21%。
The fermentation conditions and medium of Bacillus coagulans Lys 274 were optimized by single factor experiment and orthogonal experiment,and the biomass and spore number of Bacillus coagulans Lys 274 were improved.The results showed that the optimal medium composition was corn starch 10 g/L,glucose 2 g/L,lactose 5 g/L,yeast extract 20 g/L,peptone 15 g/L,beef extract 5 g/L,monopotassium phosphate 4 g/L,manganese sulfate 0.2 g/L,ferrous chloride 0.4 g/L,and the culture conditions were 50 mL liquid volume,pH 6.0,and shaking culture at 37℃for 44 h.The biomass reached 5.60×10^(9) CFU/mL,the number of spores reached 4.47×10^(9) CFU/mL,and the spore rate was 79.21%.
